补肾活血方是补肾活血法治疗糖尿病ED的有效方剂,PI3K-AKT-Caspase通路是阴茎海绵体平滑肌细胞凋亡调控的重要机制之一。前期研究表明补肾活血方可通过降低平滑肌细胞凋亡进而改善勃起功能。本课题以“PI3K-AKT-Caspase通路”为切入点,依据“PI3K-AKT-Caspase通路调控-平滑肌细胞凋亡-勃起功能” 密切关联为理论基础,提出“补肾活血方可通过PI3K-AKT-Caspase通路降低平滑肌细胞凋亡进而改善勃起功能”的假说。本研究采用补肾活血方对肾虚血瘀证生精障碍大鼠模型、原代培养大鼠海绵体平滑肌细胞进行干预,通过细胞、分子生物学和血清药理学等方法观察PI3K-AKT-Caspase通路、平滑肌细胞凋亡及勃起功能的变化,对从“PI3K-AKT-Caspase通路”揭示补肾活血方改善勃起功能机理具有重要意义。

Bushenhuoxue formula is an effective prescription for the treatment of ED in Diabetic Patients using Bushenhuoxue(Invigorate Kidney and Improves Blood Stasis)theory. PI3K-AKT-Caspase pathway is one of the most important mechanisms that regulate the apoptosis of corpus cavernosum smooth muscle cells. Previous studies show that bushenhuoxue formula can improve erectile function by reducing the apoptosis of corpus cavernosum smooth muscle cells. Therefore, we propose the hypothesis: “Bushenhuoxue formula acts through PI3K-AKT-Caspase pathway which can reduce the apoptosis of corpus cavernosum smooth muscle cells and improves erectile function.” By using bushenhuoxue formula, the study intervenes rats with impaired spermatogenesis and shenxuxueyu syndrome, also on cultured primary corpus cavernosum smooth muscle cells. We observe the changes in PI3K-AKT-Caspase pathway, apoptosis of smooth muscle cells and erectile function by cellular,.molecular level and Serum pharmacology. And the study is important for revealing the mechanism that bushenhuoxue formula can improve erectile function through “PI3K-AKT-Caspase pathway”.
