香果树(Emmenopterys henryi Oliv)隶属茜草科(Rubiaceae)香果树属(Emmenopterys),为中国特有单种属孑遗植物,具有很高的观赏及科研价值。受气候变化及人为因素等影响,该物种种群数量日趋减少、濒于灭绝,建立和完善其离体保存体系具有重要的战略意义。本项目随机选取江西省庐山、井冈山和武夷山自然保护区三个居群成年植株休眠芽构建试管苗茎尖为试材,通过玻璃化法、包埋玻璃化法和滴冻法超低温保存,探究和比较预处理方式,预培养蔗糖浓度,玻璃化液浓度及处理时间对不同保存体系下香果树再生率的影响,优化其茎尖超低温保存体系;并采用SRAP标记对超低温保存后200棵再生植株及保存过程中每步处理茎尖及培养物进行遗传稳定性鉴定,以探讨其所构建的超低温保存体系是否会影响再生植株遗传稳定性或影响遗传稳定性关键步骤,为我国珍稀濒危植物香果树有效保存提供理论和技术支持。

Emmenopterys henryi Oliv., the only species of the genus Emmenopterys belonging to the family Rubiaceae, is a ancient relic species and is indigenous to China. It has high ornamental and scientific value. However, they are now on the verge of extinction due to human impact and lack of protection. Therefore, establishing and improveing conservation system of E. henryi Oliv. has very important strategic significance. Dormant buds of E. henryi Oliv. were collected from three populations of Lushan National Nature Reserve, Wuyi Mountain National Nature Reserve, jinggang Mountain National Nature Reserve. The factors were studied that effected the cryopreservation of E. henryi Oliv. in vitro shoot tips by vitrification, encapsulation-vitrification and droplet-vitrification, such as cold hardening method, concentration of preculture, the time and concentration of PVS. Genetic stability analysis of E. henryi Oliv. before and after different stages of droplet-vitrification protocol for cryopreservation using SRAP molecular marker in the project, get a general cryopreservation protocol of this species, So the results would provide theorized and technical support for the effective way of invitro conservation of the rare and endangered species E. henryi Oliv. germplasm.
