小续命汤是祛风通络法治疗中风病的代表方剂,大量临床和基础研究已证实其确切的疗效,但均未触及其核心机制---“通络”。在总结中医络病理论和神经病学研究新进展的基础上,我们提出胶质血管单元是脑络的物质基础,小续命汤可能以细胞因子为媒介调控星形胶质细胞、血管内皮细胞交互对话,抑制急性缺血级联反应,保护胶质血管单元结构和功能,进而改善神经元生存微环境,促进神经血管单元机能重建。以此假说为基础,我们拟采用小续命汤对体内外脑缺血损伤模型进行干预,采用高通量芯片技术获取缺血后细胞因子动态变化的宏观数据,利用计算机模拟和系统分析技术对宏观数据进行处理,绘制基于细胞因子的血管内皮细胞-星形胶质细胞交互对话网络图谱,筛选其中的核心细胞因子如IL-6及其信号转导通路如PI3K/AKT、JAK2/STAT3等加以验证。通过微小环境内的大数据研究,阐明小续命汤的“通络”机制,丰富中医络病理论的科学内涵。

Dose of research indicated the effects of Xiao-Xu-Ming Decotion, however, didn't touched it's core mechanism of collateral dredging effects. Here, we propose that glia-vascular unit is the material base of brain collaterals. Xiao-Xu-Ming Decotion regulates the vascular-glia crosstalk via cytokines, inhitits the cascade after ischemia, protects structure and function of glia-vascular unit, ameliorates microenvironment of neuron, redintegrate the function of neurovascular unit. Thus, we use Xiao-Xu-Ming Decotion to treat the in vivo and vitro model of cerebral ischemia, the data of cytokine after ischemia will be detected by high-throughput array and analysed by computer simulation and system analysis technique, plot the network map of vascular-glia crosstalk, screen the core cytokines including IL-6 and its pathways including PI3K/AKT、JAK2/STAT3. This research will clarify the mecanisms of collateral dredging effects of Xiao-Xu-Ming Decotion, and enrich the theory of collateral disease.
