雌激素受体除了在其靶组织卵巢、子宫、乳腺表达外,在人体心血管系统、中枢神经系统等广泛表达,并具有重要的生理及病理意义。然而关于雌激素受体在人体小肠的表达及功能作用,国内外很少有研究。我们最近的研究发现雌激素受体在人的小肠粘膜上皮细胞表达,而且雌激素受体在小肠粘膜上皮细胞的表达分布与在其靶组织卵巢、子宫、乳腺以及胃肠道的胃粘膜上皮细胞的表达分布不同,提示雌激素受体在小肠粘膜上皮细胞具有不同的功能作用特性和不同的发挥作用的细胞分子机制。因此,在本项目中,我们主要研究雌激素受体在小肠粘膜上皮细胞生理功能调节中的作用,并了解是否因雌激素受体在胃和小肠粘膜上皮细胞的表达差异导致了其对胃及小肠粘膜上皮细胞功能的不同影响。本研究有助于阐明雌激素受体在小肠粘膜上皮细胞生理功能调节中的作用,为临床上一些疾病和生理现象发生的性别差异以及临床上开发雌激素受体调节剂对一些疾病的治疗提供理论依据。

Estrogen receptors (ERs) are widely expressed in the human cardiovascular system, central nervous system, and so on, in addition to the expressions in the reproductive organs, ovary, uterus, and breast, which mediate many biologic effects, with important physiologic and pathologic significance. However, the expression and functional role of ERs in the human small intestine are unknown. Our recent study found that ERα and ERβ are expressed in the villous cells and crypt cells of human intestinal mucosal epithelium. Moreover, the expression and distribution of ERs in the human intestinal mucosal epithelial cells are different from those in the human reproductive organs, ovary, uterus, and breast, and human gastric mucosal epithelial cells, demonstrating that ERs in intestinal mucosal epithelial cells have different functional role and different molecular and cellular mechanisms whereby ERs exert functional roles. Therefore, in the present study, we focus on studying the role of ERs in the regulation of physiological function of intestinal mucosal epithelial cells and whether ERs exert different effect on functions of intestinal and gastric mucosal epithelial cells. This study contributes to elucidate the functional role of the expressed ERs in intestinal mucosal epithelial cells and provide theoretical basis for the gender difference of prevalence of some diseases and clinically developing ER modulators for the treatment of some diseases.

雌激素受体除了在其靶组织卵巢、子宫、乳腺表达外,在人体的心血管系统、中枢神经系统等广泛表达,表明雌激素受体在人体具有重要的生理及病理意义。然而关于雌激素受体在人的小肠的表达和功能作用不清楚。在本项目中,我们研究了雌激素受体在小肠粘膜的表达及其可能的功能作用。 我们的研究表明(1)雌激素受体ERα和ERβ在人和小鼠的十二指肠和空肠粘膜的上皮细胞表达,与其在卵巢和乳腺上皮细胞的的表达分布不同,ERα和ERβ主要位于十二指肠和空肠粘膜上皮细胞的细胞浆;(2)雌激素增加了小鼠十二指肠粘膜对钙的吸收能力,雌激素受体拮抗剂ICI182.780抑制了雌激素介导的钙吸收增加。进一步的结果显示,雌激素上调了十二指肠上皮细胞的钙转运蛋白TRPV6和Calbindin-9k (CaBP-9k)的表达。 这些结果表明雌激素受体存在于小肠粘膜上皮细胞,调节了十二指肠对钙的吸收。本研究有助于阐明雌激素受体在小肠粘膜上皮细胞生理功能调节中的作用
