男性不育是一种严重威胁人类健康的男性疾病,精子活力低下是男性不育的重要原因之一,多由生殖道感染、内分泌失调、理化和药物因素所致。解脲脲原体(UU)感染所致精子活力低下的发病机制与治疗仍然是医学界面临的难题,防治本类疾病具有非常重要的意义。本研究采用出自《医宗金鉴》的知柏地黄丸为施加因素,以UU感染建立动物及细胞模型为受试对象,运用流式细胞术、Western-blot、RT-PCR、高效液相色谱法等技术,研究知柏地黄丸提高UU感染大鼠精子活力是否通过调控精子线粒体通透性转化,稳定线粒体膜电位,影响线粒体能量代谢,探讨知柏地黄丸干预UU感染对生精细胞线粒体能量代谢的影响与精子质量指标变化的相关性。从基因转录与蛋白表达水平,阐明UU感染所致男性不育的部分机制,为知柏地黄丸治疗UU感染所致精子活力低下提供实验与理论依据。

Male infertility, which is a serious disease threat to human health, is mainly caused by the low sperm motility, and the infection of reproductive infection, endocrine disorders, physical and chemical and drug factors are the main reason for the low sperm motility. Pathogenesis and treatment of low sperm motility caused by infection of Ureaplasma urealyticum (UU) is still the problem facing the medical profession, and the prevention and treatment of this disease is significance. This study uses an famous prescription named ZhiBai DiHuang Pill (which is from "Yi Zong Jin Jian") exerting factors, with UU infection animal and cell model was established for subjects by flow cytometry, Western blot, RT-PCR, high performance liquid phase chromatography technology, research ZhiBai DiHuang Pill improve UU infection rat sperm motility by regulating sperm mitochondrial permeability conversion, stabilization of mitochondrial membrane potential, mitochondrial energy metabolism of ZhiBai DiHuang Pill intervention UU infection of spermatogenic cells, mitochondrial energy metabolism effects of sperm quality changes correlation. From the gene transcription and protein expression levels and elucidate the mechanism of male infertility caused by UU infection and ZhiBai DiHuang Pill in the treatment of UU infection caused by low sperm motility provide experimental and theoretical foundation.
