糖尿病周围神经病变(DPN)是一种发病率高、危害大、病因复杂的疾病,其发病机制尚不十分明确,且无特效治疗药物。课题组前期发现,调脏通络电针治疗DPN疗效确切,并能改善DPN大鼠尾神经组织病理和传导功能。最新研究证实,内质网应激(ERS)介导的周围神经细胞凋亡与DPN发病关系紧密,为调脏通络电针治疗DPN的效应机制研究提供了新的思路。因此,课题组提出“调脏通络电针通过干预ERS介导的周围神经细胞凋亡,继而保护神经细胞并治疗DPN”的假说,计划以DPN大鼠为研究对象,利用TUNEL染色、免疫荧光、激光共聚焦扫描与三维重建等技术,观察调脏通络电针对ERS介导的周围神经细胞凋亡比例、周围神经细胞形态和表皮神经纤维密度的影响,并分析上述变化与ERS水平的关系。研究结果既可深入揭示调脏通络电针治疗DPN的效应机制,也能为DPN的防治提供新的策略和依据。

DPN is a high incidence of large harm disease with complex etiological factor, which pathogenesis is not very clear, and no specific treatment. Our previous discovery, showed that adjusting Zang organs and clearing out collaterals with electric acupuncture had therapeutic effects indeed, and can improve pathology and conduction of DPN rat tail nerve. New research confirms that has close relationship between the incidence of DPN and the endoplasmic reticulum stress (ERS) mediated apoptosis in peripheral nerve cells, which provides a new way of thinking to Effect mechanism of DPN treatment study by using adjusting Zang organs and clearing out collaterals with electric acupuncture. Therefore, the group proposed the hypothesis of" adjusting Zang organs and clearing out collaterals with electric acupuncture through the intervention of ERS-mediated apoptosis of peripheral nerves, and then protect nerve cells and treat DPN". It is plan to choose DPN rat as study object for utilizing those techniques such as TUNEL staining,immunofluorescence,confocal laser scanning and three-dimensional reconstruction. To observe the proportion of apoptotic cells in peripheral nerve, peripheral nerve cell morphology and density of epidermal nerve fibers with adjusting Zang organs and clearing out collaterals with electric acupuncture, and analyze the relationship between these changes with the ERS levels. The results not only deeply reveal the effect mechanisms by using adjusting Zang organs and clearing out collaterals with electric acupuncture for DPN treatment, but also provide the basis for new strategies and prevention of DPN.
