广西是我国乃至世界报春苣苔属Primulina Hance植物的分布中心,在这一区域分布了大量分布区域狭窄的稀有种,中草药大石七就是典型稀有种之一。该植物民间应用于肝胆疾病的治疗,利湿退黄和降转氨酶效果较好。但该植物尚未鉴定到种,关于其化学成分及药理学方面至今未有研究,无法有效利用。本项目就是在通过引种栽培获得大量大石七植株,发现其具有较强抗乙肝病毒作用的基础上,拟进一步根据形态显微特征结合ITS与trnL-F序列描述并命名该新分类群;采用体外HepG2 2.2.15细胞模型和测定HBV DNA的方法进行抗乙肝病毒活性追踪,运用多种色谱方法分离纯化活性成分,利用现代波谱技术及化学方法鉴定活性化合物的结构并研究其构效关系;建立鸭乙肝病毒模型,从DHBV病毒载量、肝功能指标、病理学检查等多环节深入探讨大石七提取物及活性化合物抗鸭乙肝病毒与保肝作用,为研发抗乙肝病毒创新药物提供科学依据。

Guangxi Zhuangzu Autonomous Region of southern China resides in the heart of an immense limestone terrain stretching across the Sino-Vietnamese border. Renowned for its picturesque karst landscapes, Guangxi is also one of the major biodiversity hotspots in China. Several genera of Gesneriaceae are but a few of the many species-rich and highly diversified plant taxa that are restricted to caves and crevices of various limestone formations. The majority of these limestone species are extremely rare, known only from a handful of localities. The vernacular name "Da-Shi-Qi" which is previously undescribed species, has long been used for treating HBV-infected patients in Zhuang and Han minority regions of Guangxi. The effect of decreasing ALT, AST and T-Bil is even better. However, we found it is different from all the known Primulina Hance species, comparing with all described Primulia Hance species. The concerned chemical constituents and pharmaceutical effects are quite few, which hinders further rational utilization and protection of the Chinese medicinal resources. Our investigation manifested that the ethanol extract of Da-Shi-Qi exhibited significant anti-HBV activities in vitro. Morphological and structural characteristics of the plant are examined using light and scanning electron microscopy. Their generic placement in the recently recircumscribed Primulina Hance is supported by phylogenetic analyses of DNA sequence nuclear ITS and chloroplast trnF-L intron spacer regions. Based on these, the new species is described and named. Its anti-HBV activity will be evaluated on the secretion of HBsAg, HBeAg and HBV DNA replication using HepG2 2.2.15 cell line and cytotoxicity to cells. The plant was extracted with ethanol and compounds in the chloroform and ethyl acetate fractions were isolated and purified by silica gel and Sephadex LH-20 column chromatographies, and preparative HPLC. Chemical structures elucidated by physicochemical properties and spectroscopic analysis. To study the effects of the Da-Shi-Qi extract on expression of DHBV DNA, hepatic function index and pathological changes of the duck liver, duck hepatitis B virus model was established. These studies provide important chemical substances and pharmacology for researching new drugs about anti-HBV treatment.
