通常骨形成是通过与骨吸收的联动作用来完成的,即我们所熟知的骨重建现象。项目申请人课题组于2011年报告了艾地骨化醇能够诱导不依赖于骨吸收的“迷你型塑造”骨形成现象。而且,我们的前期工作已经明确艾地骨化醇可以通过抑制骨基质内部骨细胞分泌硬骨素的方式促进不依存于骨吸收的“迷你型塑造”骨形成。本项目拟在前期工作的基础上,考虑通过使用硬骨素转基因小鼠模型和FIB-SEM三维超微结构观察等方式,进一步深层次探讨骨细胞调控不依存于骨吸收的“迷你型塑造”骨形成的作用机制。
英文摘要
Bone formation and bone resorption is a coupling process during bone remodeling which is a lifelong process where old or damaged bone is removed and new bone is formed in the skeleton. The applicant’s team have reported that eldecalcitol (ELD) could promote a type of focal bone formation that is independent of bone resorption in 2011, a process known as “mini-modeling” bone formation. Moreover, the preparatory work of the applicant found that secretion of SOST by osteocytes was significantly reduced at the site of “mini-modeling” bone formation by ELD administration. This study was designed to explore the effects of osteocytes on ELD induced “mini-modeling” bone formation by using SOST deficient mice model and FIB-SEM ultrastructural observation, and further to identify the underlying signaling mechanism.
