机体各种证候表现不是凭白无故的,无论是外在表现还是主观感受,疾病发生时表现出的各种症状,追根溯源,其本质就是机体整个内环境平衡发生了紊乱和破坏,机体寒热属性的不同,内环境的基本属性就有所不同,深层次地追溯,必有其现代医学的内在本质,这是中医临床有效性的坚实后盾和根基。肿瘤是一种渐变的、连续的、内外环境持续选择压力下长期的演变过程,机体本身的内在环境属性不同,其病程演变特征亦有所不同,在肿瘤启动时,整个恶性基质重塑的特征亦会不同,表观遗传机制决定了肿瘤启动时机体恶性基质的重塑特征。本项拟在寒热不同证候表观的宿体基础之上,采用微剂量缓慢造模的方式,观察这一过程中,寒、热不同内微环境宿体在面对肿瘤发生时,调节机体内稳态精确平衡与重塑的表观遗传重要机制:组蛋白翻译后修饰特征的演变特征及彼此间的差异,为临床预防、治疗,以及针对不同证候特征的患者的药物开发与研究奠定基础。

Various syndromes, no matter external appearance or subjective sensation, are resulted from deregulated balance of tissue microenvironment in essence. Basic parameters in microenvironment are varied with the body nature revealing cold- and heat-syndrome leading to a notion that internal essence of modern medicine is involved in this variation, which is considered to be robust supporter and foundation for effectiveness of Traditional Chinese Medicine (TCM) in clinic. Neoplasm is a process with gradual and persistent evolution and driven by chronic selective stimulation within internal and external environment, as well as the evolution is varied with the internal tissue microenvironment in organism. At the stage of cancer initiation, the altered characteristics of malignant matrix remodeling exist in tissues and the alteration is determined by epigenetic mechanisms. This project aimed to explore an important epigenetic mechanism regulating balance and remodeling of homeostasis during colorectal carcinogenesis induced by gentle micro-dosage in animals with cold- and heat-syndrome, and the mechanism is namely involved in the evolution profiles of post-translational modifications of histone and the difference between those modifications, providing the foundation for clinic precaution, therapy, and drug development and research in the patients with cold- and heat-syndrome.
