棉花的生殖器官蕾花铃作为棉铃虫危害的首选目标,研究蛋白质合成及分解的外源调节对Bt转基因棉生殖器官Bt杀虫蛋白表达量影响程度及原因对于解决Bt棉存在的花铃期及生殖器官抗虫性低等问题具有重要意义。本项目拟通过增加土壤氮素用量、生殖器官应用尿素、氨酸酸组分及联合应用等促进蛋白质合成;土壤氮素亏缺、生殖器官应用氨氧乙基甘氨酸(AOA)、蛋氨酸亚砜亚胺(MSO) 等降低蛋白质合成;生殖器官应用EDTA、丝氨酸蛋白酶抑制剂(PMSF)等抑制蛋白质分解、应用Mg2+、Ca2+等促进蛋白质分解等处理探讨对Bt杀虫蛋白表达量影响及其蛋白质周转生理机制,明确生殖器官蕾花铃Bt杀虫蛋白表达量的可调程度及原因,为寻求实现Bt棉产量器官抗虫性提高的途径提供相关的栽培生理基础。

The reproductive organs are first targets when the boll worm harm cotton, studying the effect and mechanism by external regulation of protein synthesis and degradation on the insecticidal protein expression of the reproductive organs is important to solve the low insect resistant expression at flowering and boll setting periods and at reproductive organs in Bt cotton. The enhanced protein synthesis treatments by nitrogen application, urea and amino acid composition spraying, the reduced protein synthesis treatments by nitrogen dificiciency, AOA and MSO application, reduced protein degradation treatment by EDTA and RMSF, increaded protein degradation treatment by Mg2+、Ca2+ application on the reproductive organs will be conducted to investigate their effects on the Bt protein expression and physiological reason. The affected extent of insect resistance of the reproductive organs and related physiological mechanism will be probed under the different treatments above respectively, the effect of insect resistant expression will be ascertained by external redulation of the protein cycle .These investigation will provide cultural physiological basis for the approach of enhancing insect resistance of the yield organs.
