细胞治疗是近年来快速发展和极具潜力的一种治疗方法。磁共振分子影像是在体细胞追踪、定量和评估最有效的影像学工具。由于目前缺少一套既可以进行细胞标记又可以对被标记细胞进行特性分析的标准化仪器,本项目拟研制一种全自动流式细胞超声标记仪,用于对细胞进行连续磁标记和对被标记细胞进行特性分析。该系统基于超声声孔效应、用磁共振造影剂标记细胞,基于微流控技术和膜分离技术、用稀释过滤法洗涤细胞,基于惯性分离和材料磁性分选细胞及定量标记物,基于阻抗法计量和分析细胞。该系统包括流动进样、细胞超声标记、细胞洗涤、细胞分选与载磁量化、细胞计数与活性分析等子系统,用流式细胞技术实现对细胞的自动连续标记和分析,实现对不同类型细胞标记参数的自动优化和实验评估。本系统的研制成功将为磁纳米粒子标记细胞提供快速、自动、高效、安全、无菌和标准的一体化技术平台,大大推动细胞治疗的基础研究和临床应用进程。

Cell therapy is a rapid developing and promising method for treatment of various diseases. Molecular magnetic resonance imaging is the most effective tool for cell track, quantification and evaluation. Due to the lack of a standard instrument that can not only label cells but also analyze the properties of the labeled cells, this project is to develop an automatic flow cytomagnetosonoporation (FCMSP) instrument for continuously labeling cells and analyzing the properties of the labeled cells. This instrument is to label cells with MRI contrast agents based on the sonoporation effect, wash cells using a dilution-filtration method based on microfluidic and membrane techniques, sort cells and quantify agents in cells based on inertia and magnetism separation techniques, and count cells and analyze cell viability based on impedance measurement. The system will include several subsystems for flow sampling, cell labeling, cell washing, cell sorting, agent quantification, cell counting, and cell viability analysis respectively, and be able to label cells and analyze labeled cells by flow cytometry continuously and optimize the labeling parameters for different kinds of cells automatically, followed by in vitro and in vivo evaluation. The success of this project will provide a rapid, automatic, efficient, safe, sterile and standard integrated technology platform for cell labeling with magnetic nanoparticles, and will greatly promote the basic research and clinical applications of cell therapy in the near future.
