山核桃雌雄同株、童期长,雌雄花发育过程中的营养生长向生殖生长转变是花发育一个非常重要的关键点。lncRNA在调控植物发育方面发挥着重要作用。本项目基于山核桃全基因组序列、成花相关表达谱、microRNA和成花相关基因功能等方面的研究,构建山核桃雌雄花发育过程中营养生长和生殖生长阶段的4个lncRNA文库,运用高通量测序及生物信息学方法分析挖掘得到山核桃成花期的保守和新的lncRNA及其靶基因,并应用统计学方法分析雌雄花营养生长和生殖生长过程中的lncRNA表达差异,将表达差异显著的lncRNA及其靶基因转入山核桃胚胎干细胞和拟南芥中验证功能,并利用酵母单杂和ChIRP技术分析关键lncRNA与DNA和蛋白相互作用,从而解析lncRNA在山核桃成花过程的分子调控功能。
