系统性红斑狼疮患者的生存率在过去几十年有了极大提高，但大多数人的病情容易反复，有较好的一年疾病控制期的病人不到20%。中药治疗SLE可以有效的降低疾病发作率，减少糖皮质激素的使用量，解毒祛瘀滋肾方和升麻鳖甲汤是治疗SLE的有效复方并具有明确的抗炎效应，但同样存在疗效不突出的共性问题。进一步增强该类复方的抗炎作用，减少患者对糖皮质激素的依赖对于提高复方的临床疗效具有重要意义。在本课题中，我们制备糖皮质激素受体（GR）和核因子-κB (NF-kB)蛋白的亲和层析柱，利用亲和层析的方法靶向分离中药复方中的抗炎复合物。通过检测复合物对细胞中GR和NF-kB蛋白的调控以及对狼疮鼠炎症抑制作用，评判此类抗炎复合物对狼疮活动的干预机制。课题研究期望获得药物质量可控并且作用靶点明确的现代中药复方，从而为SLE的中药新治法提供思路。

The survival rate of patients with systemic lupus erythematosus (SLE) has been greatly improved in the past decades, but most of the disease is not well controlled, and it is easy to relapse in the process of drug withdrawal. One year of effective disease control is less than 20%. Traditional Chinese medicine treatment of SLE can effectively reduce the patient's disease onset and reduce the use of glucocorticoid. Jieduquyuziyin prescription (JP) and Shengmabiejia decoction (SD) are widely used for SLE treatment and shown anti-inflammatory effect. How to further enhance the anti-inflammatory effect and reduce SLE patient dependence on glucocorticoid is a very important thing to improve the efficacy of traditional Chinese medicine. In this study, we prepared the affinity chromatography column of glucocorticoid receptor (GR) and nuclear factor kappa B (NF-kB) protein, and separated the components which can bind with GR and NF-kB from JP and SD by affinity chromatography. By detecting the isolated components on the regulation of GR/ NF-kB signaling pathways and the inflammatory responses of MRL/lpr mice, we hope to screen anti-inflammatory substances and establish a new method for the treatment of SLE with Traditional Chinese Medicine. This research will lay the foundation for modern Traditional Chinese Medicine prescriptions with clear components and definite therapeutic targets.
