On the basis of our previous studies about virus infection, immune response against virus infection and development of vaccines and drugs, the project will focus on the key issue of preventing and treating Ebola virus infection to perform the research works in five aspects, including the virology, structural biology, immune response and pathogenesis, interaction between host and virus, mechanism of antiviral drugs. Through comprehensive analysis of the data including genomics, proteomics, pharmaceutics, immunology and protein structures, we will identify how the Ebola virus evolves and the cellular and molecular mechanisms of virus entry, replication and packing, and elucidate the interaction modes between host immune system and virus infection, host factors and virus proteins. In addition, we will also study the molecular mechanism of current antiviral drugs and explore new drug target. Our study will provide new clues for the creative investigation of Ebola virus infection and new mechanistic insight into the prevention and clinical treatment of Ebola virus infection, thus outlining new potential targets for the design of drugs and vaccines.

该项目在短短的两年时间里,项目取得了重大突破。在国际上率先揭示了埃博拉病毒糖蛋白结合内吞体受体NPC1的分子机制,首次提出一种新的囊膜病毒膜融合激发机制(第五种机制),为防控埃博拉病毒病疫情及抗病毒药物设计与研发提供了重要科学基础,相关工作发表在国际顶级学术期刊《Cell》上。解析了埃博拉病毒核蛋白NP的三维结构,阐明了病毒遗传物质的组装方式。揭示了埃博拉病毒表面抗原-ZMapp中和抗体相互作用机制。揭示了埃博拉病毒在宿主体内进化的规律,首次发现非编码突变对于埃博拉病毒的影响,提示病毒最保守基因VP40可以作为有效的抗病毒靶点。筛选获得了具有自主知识产权的3个高亲和力中和抗体,3种多肽类药物,和 10多个小分子候选药物,还进一步研发了组合药物和双特异性抗体。优化现有抗体药物MIL77的表达和联合使用策略,获得国家CFDA临床批件。成功研发了多种埃博拉病毒检测试剂及配套仪器,通过国际第三方评价并用于非洲地区的样本检测。这些工作为防治埃博拉病毒病提供了坚实的理论基础和强有力的技术支撑。该项目发表论文49篇,其中SCI论文44篇,影响因子大于10的论文7篇(包括2篇Cell,1篇Immunity);申请专利13项,申请临床批件4项。超额完成研究任务。
