激素性股骨头坏死(SIONFH)是长期或大量应用激素导致股骨头内活性成分死亡的病理过程,目前发病机制尚不明确且诊疗方法局限。研究表明,转录组学涵盖的基因转录调控机制或对骨坏死的病变发展与修复起重要作用。SIONFH在转录环节存在关键基因网络可调控骨代谢、脂化及血管化失衡或再平衡,包括成骨、破骨、成脂化、血管化等信号通路的基因或蛋白。本研究拟采用高通量RNA-seq技术进行全转录组测序,全面快速地获取特定状态下组织内所有差异基因的表达信息;结合基因沉默技术研究差异基因与相关信号通路之间的关系;进一步探究“祛瘀法”对疾病发展过程中坏死组织转录组基因的调控机制。试图从转录组学角度研究SIONFH的病变发展,探讨“祛瘀法”促进SIONFH修复的作用机制,为中药防治SIONFH的作用靶点研究提供新思路。

Steroid-induced osteonecrosis of femoral head (SIONFH) is the pathological process of the bone tissues necrosis due to the frequent or long-term application of glucocorticoid. Until now, pathogenesis of the disease is remained unknown and treatments limited. Recent studies showed, transcriptomics regulation play an important role in development and repair of SIONFH. There are several key gene networks exiting in the transcription of SIONFH for regulating the imbalance or rebalance of bone metabolism, adipogenesis and angiogenesis. The networks cover key genes or proteins in particular signal pathways, includes osteogenesis ones, adipogenic ones, angiogenesis ones and immune related ones. We intend to use the high-throughput RNA-seq analysis for sequencing transcriptomes; obtain the information of all different transcripts (DT) in full range quickly; discover the relationships between the DT and related signal pathways by siRNA technique; verify the transcriptomics regulation of “removing blood stasis” method in disease development. All of these are practical for us to evaluate the mechanism of the disease, of “removing blood stasis” method, and provide a creative approach for drug targets selection of herbs in SIONFH.
