The applicant has been engaged in the understanding of the pathogenesis and therapeutics of myocardial injury under metabolic diseases. The applicant’s major scientific contributions encompass (1) revealing oxidative stress and lipid toxicity-initiated autophagy and novel mechanisms behind myocardial injury in metabolic disorders from the perspective of compromised protein accumulation; (2) identifying ER stress induced mitochondria damage as caspase effect and novel mechanisms of myocardial injury in metabolic disorders from the perspective of organelle damage;(3)discovery of aldehyde dehydrogenase 2 (ALDH2) governed bi-directional autophagy regulation as the upstream switch protein to promote removal of damaged proteins. ALDH2 protects myocardial proteins and organelles including endoplasmic reticulum and mitochondria from pathological insults in metabolic disorders. These findings have shed some promises towards identification of new therapeutic targets in heart disease. Over the past five years, the applicant has published 30 papers in first-rated journals as the first or corresponding author. Among them, 25 publications were published in journals with impact factor higher than 5. The combined impact factor reaches 195.4 with 280 citations from others including Physiological Reviews with IF of 29.041. The applicant has served as the principal investigator for two NSFC grants and played a major role in research awards from National Institutes of Health. The applicant was also honored as the Outstanding Researcher twice in the Center for Cardiovascular Research and Alternative Medicine at the University of Wyoming and received the prestigious awards as an excellent young technical talent in the General Logistics Department and Shannxi provience. Serving as a guest editor for journals such as Curr Pharm Design and BBA Mol Basis Disease, the applicant edited special issue focused on metabolic disorder. The applicant also serves on editorial board for Cardiovascular Diagnosis and Therapy, BioMed Research. This proposal is aimed to examine the role of ALDH2 in diabetic cardiomyopathy as control protein of mitochondrial quality control, in an effort to identify new therapeutic target for cardiac dysfunction under metabolic syndrome.
