创新性地提出以中药"动态血清化学"研究为中心,结合药代、药效快速筛选与评价技术进行中药复杂体内效应物质的确证,促进中药的深层次开发,提高我国创新中药的研发成功率与效率。在阐明体内效应物质的基础上,运用多组份同时定量分析技术,开展多组份药代动力学研究。首次提出运用药代动力学原理进行多组份药时曲线的整合从而获得能代表全方的整合药时曲线与参数。率先应用代谢组学研究思路与技术,建立中药方剂整体药效评价体系,并运用模式识别等确定客观量化的整体药效评价指标,进而结合药理效应法的研究思路建立"代谢组效应法"整体药代动力学评价以及PK-PD 研究体系,从而获得具有切实指导意义的整体药代及药效参数,指导临床合理用药,为中药复杂体系的现代化研究开辟全新的研究思路与方法。
英文摘要
In order to promote the deep reseach and exploration of TCM and to enhance the efficacy and power of developing novel TCM, we creatively brought forward in the present project a systematic research idea and method concerning the integrated pharmacokinetic and pharmacological reseach on TCM.Combined pharmacy and serum chemistry, pharmacoloigal and pharmacokinetic sreen method will be adopted to discover the pharmacological materials of, and the potential new drug candidates from the TCM complication. A new concept of integrated pharmacokinetics has been clearly defined as modeling the multi-components plasma profiles of TCM to obtain an integrated concentration-time curve and thus integrated pharmacokinetic parameters representing the whole TCM.Metabonomics, as the biochemial phenotype of the whole body function, will be introducted into establishing the novel pharmacological evaluation system for the TCM complicated activities. Furthermore, a dynamic metabonomic strategy including dose and time dependent study will be used for the integrated pharmacokinetic and PK-PD evaluation as the idea used in the traditional biological effect method for TCM pharmacokinetics evaluation. The creative idea adopted in the project will lead to the deep and novel acknowledge on the pharmacokinetics and pharmacological activities
