胰岛素样生长因子是一类对骨形成和吸收具有重要调节作用的生长因子,本研究采用PCR-RFLP技术和mRNA定量技术对鸡IGFs基因进行分型,研究不同品种的健康和骨质疏松患鸡IGFs基因的多态性分布情况及其对IGF轴的影响,探讨IGDFs基因多态性与骨密度的关系,从基因水平了解该病机理,为抗骨质疏松品种选育和防治提供理论依据。

90、180 hens of ISA and white leghorn were chosen randomly. All samples' IGF-Ⅰgene fragments mapped in the site of 7kb of promoter upstream were detected by means of PCR-RFLP, the gene fragments were digested with PstⅠand the relationship between genetic polymorphism and bone quality, bone biochemical marker and body weight etc were analysed. Result showed that the frequencies of PP, Pp, pp genetype were 0.256、0.311、0.433 in ISA layer, respectively. With statistical analysis (ANOVA), it was showed that the BMD and BMC of humerus of PP genetype was obviously higher than these of Pp, pp genetype, The body weight of PP genetype was heavier than these of Pp pp genetype. While the frequencies of PP, Pp, pp genetype were 0.028、0.061、0.911 in white leghorn layer, respectively. Statistical analysis showed humerus radiographic density, cortical width, tibia cortical width and bone volume of genetype of pp were significantly higher than these of the other genetypes, there was certain relationship between BGP、AKP and IGF-Ⅰgenetype, the concentration of serum BGP、AKP of genetype pp(Pst1+/+) was obviously higher than these of the other genetypes. 随机选取ISA、white leghorn蛋鸡各90、180羽,应用PCR扩增鸡IGF-Ⅰ启动子上游7kb处基因片断,PstⅠ酶切消化产生限制性片断长度多态性,分析IGF-Ⅰ多态性和骨质,骨代谢标志物、体重等之间的关系。结果:ISA蛋鸡PP、Pp、pp基因型频率为0.256、0.311、0.433,方差分析显示肱骨骨密度和骨矿含量PP基因型均显著大于pp、Pp基因型;PP基因型体重显著大于Pp基因型。而white leghorn蛋鸡PP、Pp、pp基因型频率分别为0.028、0.061、0.911。方差分析显示肱骨放射骨密度、皮质骨厚度、胫骨中段皮质骨厚度及骨小梁体积pp基因型均显著大于Pp、PP基因型;骨钙素、碱性磷酸酶pp基因型显著大于PP、Pp基因型。结论:笼养蛋鸡IGF-Ⅰ基因多态性和骨质显著相关,较大体重有利于笼养蛋鸡骨质发展。ISA笼养蛋鸡P(Pst1-)等位基因具有一定的优良骨质指针作用,而white leghorn笼养蛋鸡p(Pst1+)等位基因具有一定的优良骨质指针作用,提示从分之水平探讨骨质丢失对进一步深入研究笼养蛋鸡抗病基因选育有重要实际意义。
