夏玉米收获期籽粒含水率较高是限制黄淮海玉米机收籽粒进程的主要瓶颈。夏玉米生理成熟后田间站杆时间短,无法完成籽粒物理脱过程,是导致收获期含水率高的直接原因,开展生理脱水过程相关生理机制研究对生产具有重要意义。本项目以玉米籽粒生理脱水过程为研究对象,在2014-2016年工作基础上,以生理脱水差异较大的2组(4个)玉米杂交种为材料,以脱落酸所介导的玉米籽粒生理脱水机制为切入点,运用外源ABA与ABA合成、水通道抑制剂喷施相结合的方法,在籽粒灌浆特征和生理脱水规律等研究基础上,运用双向电泳、膜片钳全细胞记录技术等研究手段,系统开展脱落酸含量变化规律、与其他激素互作关系、比较蛋白质组学、胚乳细胞水通道调控等方面的研究。以期明确脱落酸介导的籽粒生理脱水机制和调控机理,为降低生理成熟期籽粒含水率,实现玉米机收籽粒和品质提升提供理论基础,同时也可为玉米脱水相关生理基础性研究及玉米育种提供全新的思路指导。

Harvest of summer maize grain with high water content is the main bottleneck in the process of harvesting corn grain. After physiological maturity of summer maize field station pole time short, unable to complete the grain physical removal process is a direct cause of the harvest period with high moisture content, carry out the relevant research of physiological mechanism physiological dehydration process has important significance for the production. This project is based on the maize physiological dehydration process as the research object, on the basis of 2014-2016 years of work, with 2 sets of large differences in physiological dehydration corn hybrids(4 varieties) with abscisic acid mediated maize physiological dehydration mechanism as the starting point, using the method of exogenous ABA and ABA synthesis, water channel inhibitor spraying combined, based on Grain Filling Characteristics and physiological dehydration law research, using two-dimensional electrophoresis, whole cell patch clamp recording technique and other research methods. The changes of abscisic acid content, the interaction with other hormones, the comparative proteomics, and the regulation of water channel in endosperm cells were systematically studied. In order to clear off the physiological mechanism and regulation mechanism of seed dehydration induced by acid, to reduce the physiological maturity of the grain moisture content, provide a theoretical basis for maize grain harvesting and quality promotion, but also provide the physiological basis of research and guidance for maize breeding new corn dehydration.
