钉螺孳生导致血吸虫病的流行难以控制,严重制约着血吸虫病流行区的经济发展。申请者前期研究发现,外来入侵物种加拿大一枝黄花(Solidago canadensis)提取物通过影响钉螺体内酶的活性和破坏肝脏等器官的结构而对钉螺产生较强的杀灭作用,杀灭效果呈现浓度和时间依赖性,48h的IC50为0.57mg/mL。申请者拟在此工作基础上,采用活性追踪分离技术确定加拿大一枝黄花提取物的灭螺活性组分;采用GC-MS、MALDI-TOF MS、HPLC-ESI-MS技术对活性部位中的已知化合物进行快速指认;综合应用色谱、波谱技术定向追踪分离、鉴定具有抑螺活性的新型单体化合物;通过检测活性组分和活性化合物对钉螺超微结构以及蛋白质和糖原含量、酶促动力学等的影响来探讨抑螺活性的作用机理,评价活性组分和化合物的灭螺活性,为将加拿大一枝黄花用于钉螺防治、开发新型灭螺剂提供依据,实现对加拿大一枝黄花的高值资源化利用。

Oncomelania hupensis is the only intermediate host of schistosome.O. hupensis breeding in Poyang Lake wetland causes schistosomiasis epidemic is difficult to control, seriously restricting economic development in the schistosomiasis area . Previous research indicated that the extracts of the alien invasion species-Solidago canadensis have strong killing effect on O. hupensis with IC50 0.57mg/mL withing 48 hrs by damaging liver anatomical structure and affecting activities of some enzymes, showing concentration and time dependent. On the base of these works, Applicants will identified known compounds in the extracts of S. canadensis by means of GC-MS, MALDI-TOF MS, HPLC-ESI-MS rapidly, isolate and purify the constituents that have the inhibition activity on the O. hupensis in S. canadensis by means of column chromatography such as Si Gel, ODS, Shaphadex LH-20 guided by bioactivity screening. The structures of the constituents will be elucidated on the basis of spectral analysis and chemical derivation. The inhibitory activity of the constituents will be detected by biochemistry methods. By means of these research works, we hope to find some inhibitor leading compounds. In this study project, the molluscicidal-activities of the extraction, separation and identified active compounds were systematically investigated, and the inhibition mechanisms were tested based on biochemical and ultra structural works. We believe this work can provide new method for scientific evaluation and rational use of molluscicidal plants.
