申请人在前期研究中将硒引入到铜(II)配合物的分子结构中得到含硒铜(II)配合物,研究发现铜(II)配合物和含硒铜(II)配合物均能富集在小鼠肝脏;均能有效抑制肝癌细胞的增殖,诱导肝癌细胞凋亡,且对正常细胞毒性较低;基于以上初步观察事实的比较,发现含硒铜(II)配合物优于不含硒铜(II)配合物;但含硒铜(II)配合物能否发挥普通有机硒的免疫调节功能,是否比结构相似的铜(II)配合物有更优越的抗癌效果,尚未得到更多的事实依据和充分阐明。本课题将进一步设计合成含硒铜(II)配合物,并以结构相似的不含硒的铜(II)配合物作为对照,系统研究阐明含硒铜(II)配合物抑制肝癌MHCC97H细胞侵袭转移作用及其分子机制;阐明硒的引入对含硒铜(II)配合物的功能增强作用。为新型抗肝癌金属药物的设计合成提供理论和实验依据,为含硒铜(II)配合物的临床前应用研究奠定基础。

This study introduced the selenium element in the structure of Copper(II) complex, it is found that Copper(II) complex with selenium and without selenium both exhibited great inhibition against liver cancer MHCC97H cells proliferation and low toxicity to normal cells, and induced apoptosis of liver cancer cells. Based on above researches, it is suggested that Copper(II) complex containing selenium displayed better bioactivity than without selenium; but it is still unclear if Copper(II) complex containing selenium can drive similar immune regulatory function of organic selenium compounds, and if Copper(II) complex containing selenium exhibited better anti-cancer effect than similar structure of Copper(II) complex without selenium. This study further design and synthesize a series of Copper(II) complex containing selenium, compared with Copper(II) complex without selenium, in order to clarify its inhibition and mechanism of invasion and metastasis of liver cancer MHCC97H cells blocked by Copper(II) complex without selenium; and illustrated if the introduction of selenium can enhance the anti-cancer activity of Copper(II) complex. For the design of novel metal complexes inhibited liver cancer provides theoretical and practical basis, and lay the foundation for pre-clinic application trials of Copper(II) complex containing selenium.
