黄精是传统大宗中药材,其中滇黄精Polygonatum Kingianum Coll. et Hemsl.是中药黄精的主要来源之一。黄精多糖是黄精中主要的活性成分,也是影响黄精品质的指标性成分,本项目拟开展滇黄精多糖代谢关键酶基因及其调控糖积累机制的系统研究。基于前期研究基础,以滇黄精为试验材料,设计不同发育时期、不同种质资源、不同蔗糖、不同光照和干旱胁迫等试验。应用气相色谱(GC)法测定滇黄精根茎多糖含量和组成,并分析糖代谢相关酶的活性变化。通过上述协同分析,掌握滇黄精根茎多糖含量和组成的变化特征、掌握滇黄精根茎糖代谢酶活性差异,揭示滇黄精根茎多糖含量和组成与糖代谢酶活性的相关关系,进一步发掘并获得滇黄精多糖代谢关键酶。明确滇黄精多糖积累差异的生理机制,为进一步阐明滇黄精品质形成和调控机制提供理论依据。

The Polygonatum kingianum Coll. et Hemsl. is the main source of the Polygonati Rhizoma, which is an important traditional Chinese medicine. The polysaccharides is main active components as well as a key content to appraise the quality of the medicine. Based on the result of the former studies, the contents of polysaccharides and activity of sucrose metabolism-related enzyme need to be determined using by the GC method and enzymes activities analysis, including the five experiments in the different rhizome development stages, germplasm resource, sugar contents, light and drought stress. To find the key polysaccharides metabolism-related enzyme based on the polysaccharides dates, and the difference characteristic of sucrose metabolism-related enzyme, the correlations between polysaccharides changes and sucrose metabolism-related enzyme activities are analyzed in the five experiments. It will define the physiology mechanism during the sugar accumulation stages of the P. kingianum, and provide the theory basis for the polysaccharides regulative mechanism and the quality control of the Polygonati Rhizoma.
