微管是两种在植物细胞生命活动中起重要作用的细胞骨架结构之一,其通过高度的动态变化参与细胞生命活动。细胞在增殖过程中,微管在不同列阵间转换并受微管结合蛋白(MAPs)的调节。由于人们对微管列阵间转换的调控机制的了解仍十分有限,因此发现新MAPs并对其功能进行研究一直是微管骨架研究领域的重点内容之一。本实验室在挖掘新微管结合蛋白时,筛选获得了一个可能含有多个功能结构域的微管结合蛋白(MAP96)。将其在细胞中表达后发现,该蛋白与细胞分裂进程及细胞扩大生长过程中不同微管列阵分布模式相似,并且药理学研究结果显示其主要与微管骨架系统相关,暗示MAP96可能是一种重要的微管结合蛋白。本申请拟在前期工作基础上,利用遗传学、细胞生物学、生物化学等学科手段对MAP96的生化特性及其在细胞中的功能,尤其是在细胞增殖过程中的功能进行研究。预期研究结果为深入认识细胞增殖过程中微管列阵动态转换的调控机制奠定基础。

The microtubule cytoskeleton, one of cytoskeletal structures that are highly dynamic changes in plants, plays a significant role in cellular activities, especially in cell proliferation. Microtubules convert during different arrays and are regulated by microtubule-associated proteins (MAPs). The molecular mechanism in converting microtubule arrays is less known, hence exploring new MAPs and their functionally studies are very important in the field of the microtubule cytoskeleton. In our laboratory, we screened a possible microtubule-associated protein (AtMAP96) which contain multiple domains from the Arabidopsis database. Through using eukaryotic expression system, we found that the distribution patterns of AtMAP96 are similar as different microtubule arrays during cell division progression and cell expansion progress. Pharmacological results also indicated that AtMAP96 is mainly associated with the microtubule cytoskeleton. These data suggested that AtMAP96 may be one of the important MAPs. Therefore, we apply, based on our previous work, to study the biochemical characteristics and cellular functions of MAP96, especially in the functional study during the process of cell proliferation, by making use of these methods of genetics, cell biology, biochemistry and so on. It is expected that these results of this study will provide a new idea for the profound understanding of the dynamic regulation of microtubule structures in the process of cell proliferation.
