南方红豆杉是我国特有的濒危药用孑遗植物,其种子休眠与萌发机制尚不明确,且休眠期长,萌发率低,严重阻碍了该珍稀物种的育种进程。本项目拟采用蛋白质复合体组学方法及质谱成像技术,通过结合二者整体性和原位分析的优势特点,基于南方红豆杉休眠与萌发种子不同生理状态,针对种子内部蛋白质复合体、相互作用蛋白质、蛋白质翻译后修饰及内源性代谢小分子开展整体、动态、原位分析;通过结果整合、综合分析以期在蛋白质互作及代谢层面上全面、精确揭示南方红豆杉种子休眠与萌发机制,指导育种进程、推动优质种质资源的长期保存。本研究建立的分析方法及研究思路亦可应用于其它植物种子休眠萌发机制研究,为农林育种提供新的研究策略与分析手段。
英文摘要
Taxus chinensis var. mairei is a relict plant (living fossil plant) in our country. The mechanism of seed dormancy and germination is still unknown, and the breeding process of Taxus chinensis var. mairei is usually hindered by long dormancy of seed and low germination rate. The methods of protein complexomics and mass spectrometry imaging (MSI) are used in this study to combine both advantages of integrity analysis and in situ characterization in mechanism analysis for seed dormancy and germination of Taxus chinensis var. mairei. Based on protein complexomics and MSI, the development of a synthetic method for overall, dynamic and in situ MS analysis about comprehensive objects (protein complexes, interacting proteins, protein post-translational modifications, and endogenous metabolites) from Taxus chinensis var. mairei seeds with different physiological status between dormancy and germination will be carried out. The results integration and thorough analysis are useful for comprehensively and accurately revealing the mechanism of Taxus chinensis var. mairei seed dormancy and germination. The breeding process and storage of high quality germplasm resources of Taxus chinensis var. mairei are also benefited from this project. The establishment of this comprehensive characterization method will be also significant in mechanism analysis for other rare plants' seed dormancy and germination, providing a new research strategy for agroforestry.
