呼吸道合胞病毒(RSV)是引起婴幼儿呼吸道疾病发病和致死的重要感染源。现有医药技术在防治RSV感染方面还十分薄弱,研制开发新型抗RSV药物刻不容缓。RSV M2-1蛋白是病毒复制过程重要的转录延长/终止抑制因子,控制着病毒在宿主细胞内的复制和转录过程。作用于RSV M2-1蛋白的小分析抑制剂能够阻断病毒在宿主内复制而发挥抗病毒作用,是新型抗RSV病毒研发的重要途径和方向。在前期工作中对枳椇属枳椇子中的黄酮碳苷类成分进行抗RSV活性研究,发现该类化合物能够抑制多种RSV病毒,在病毒进入宿主后的复制阶段发挥作用。本项目拟在前期工作基础上,进一步对枳椇子黄酮碳苷类成分进行分离和鉴定,评价其对抗RSV病毒活性,研究其作用机制和构效关系,为新型高效安全的抗RSV病毒药物的研发提供科学依据。

Respiratory syncytial virus (RSV) is an important morbidity and mortality factor on respiratory disease in infants. The existing medical technology for the prevention and treatment of RSV infection is still very weak, and developing new anti -RSV drug is urgent. RSV M2-1 protein is an important transcriptional antiterminator in the process of viral replication in the host cell. It is an important to develop new type RSV inhibitors acting on M2-1 proteins. In the previous work, we found that the flavonoid C-glycosides from Hovenia acerba Lindl. could inhibit many RSV virus, playing anti RSV activity in the replication stage after the virus enters the host. In this project, more flavonoid C-glycosides will be separated and identificated from H. acerba, the anti RSV activity will be evaluated and the mechanism and structure-activity relationship will also be further studied. The project will provide a scientific basis for the development of new efficient and safe anti viral drugs RSV.
