草珊瑚(Sarcandra glabra)是中国传统中草药,已经研制成各种药剂临床上用于治疗各类炎症,但是草珊瑚的种质资源信息还很不完善,尤其是存在与海南草珊瑚(S. hainanensis)混淆的现象。本研究拟对草珊瑚属两种分布重叠的云南、广西、广东和海南等地至少8个居群采样,通过转录组测序的方法,开发出高通量的SNP(单核苷酸多态性)分型芯片。利用这些芯片产生的SNP分型信息评估草珊瑚属两个物种的遗传多样性,并通过相关性分析找到与药物活性成分含量相关的SNP,追溯其对应的转录本功能,为优良品种的选育提供参考。最后从SNP分型信息中找到在两个物种之间稳定分化的SNP,用于草珊瑚属的分子鉴别,并评价其鉴别效力。

Sarcandra glabra is traditional Chinese medicine, which has been developed into numerous drugs to heal different kinds of inflammation. However, germplasm resource information of S. glabra is still incomplete. Especially, S. hainanensis is sometimes mistaken as S. glabra. This study aims to sample many individuals from at least eight populations in the overlap distribution areas of the two species in Yunnan, Guangxi, Guangdong and Hainan, and develop high-throughput SNP genotyping array using transcriptome data. Using the resultant SNP genotyping data, we analyze genetic diversity of the two species and find the SNPs related to the level of medicine's active chemicals, review the function of corresponding transcripts and assist the selection of new cultivar. Finally, we find the SNPs which can steadily distinguish different species, and use them to identify different species and evaluate their performance.
