近年来,我国儿童肥胖和高血压患病率呈现增加趋势。儿童肥胖和高血压不仅会引起近期靶器官损害,还增加成年期心血管病风险。申请人前期工作包括:(1) 探讨了 GWAS发现的肥胖易感基因,及其与环境因素交互作用对儿童肥胖和高血压的影响,成果发表在Am J Epidemiol和Int J Obes等;(2) 探讨了GWAS发现的高血压易感基因,及其与肥胖的交互作用对儿童高血压的影响,成果发表在J Hum Hypertens和Am J Hypertens等;(3) 通过国际合作,首次研制出儿童高血压国际参照标准,同时提出和验证了儿童高血压标准的简化方法,成果发表在Circulation、Pediatrics和Hypertension等。申请人拟采用代谢组学方法筛选出关键生化标记物,建立儿童高血压早期筛检模型;通过系列动物实验对关键代谢标记物进行功能学验证,探讨儿童高血压发生的相关代谢通路和病因机制。

Prevalence of obesity and hypertension in Chinese children has increased during recent years. Childhood obesity and hypertension are not only associated with target organ damage, but also increase the long-term risk of cardiovascular disease in adulthood. The preliminary work performed by the applicant include: (1) Examination of the influences of obesity related genetic loci identified by GWAS and their interactions with environmental factors on risk of obesity and hypertension in Chinese children. These results have been published in SCI journals including Am J Epidemiol and Int J Obes. (2) Examination of the influences of hypertension related genetic loci identified by GWAS and their interactions with obesity on risk of hypertension in Chinese children. These results have been published in SCI journals including J Hum Hypertens and Am J Hypertens. (3) The establishment of international child blood pressure references, which has been published in Circulation. Meanwhile, the applicant proposed and validated several simplified pediatric blood pressure evaluation methods. These results have been published in Pediatrics and Hypertension. The applicant aims to develop the prediction model of childhood hypertension based on the metabolic biochemical markers identified by Metabonomics method. Meanwhile, a series of experiments will be used for the function validation of the metabolic biochemical markers, and examine the underlying metabolic pathways and mechanisms of childhood hypertension in this study.
