随着超重和肥胖人口的不断攀升,中国已成为糖尿病人口大国。《六因条辨》黄连温胆汤(温胆汤加黄连)可纠正高脂饮食合STZ腹腔注射建立T2DM大鼠血糖血脂异常,且呈量效关系。已完成国自然青年基金项目结果显示,温胆汤原方具有改善高脂饮食长期喂养诱导IGT大鼠糖脂代谢紊乱及IR,其机制与改善肝脏、骨骼肌胰岛素后信号通路有关。故本项目拟从方剂配伍角度从药学、药理学的角度筛选治疗T2DM的黄连与温胆汤最佳配伍比例,并比较该方与温胆汤原方、黄连在治疗T2DM方面的药效学差异,为进一步应用分子生物学及系统生物学技术探讨该方治疗T2DM作用机制及作用靶点奠定基础,为黄连温胆汤及其配伍治疗T2DM提供实验依据,为类方研究提供思路,也为进一步建立中医复方系统研究平台奠定基础。

With the rising population of overweight and obesity, China has become a great nation with diabetic population. Huanglian wendan decoction, out of Treatise on six reasons, (Wendan decoction adding rhizoma coptidis ) can amend the blood glucose and lipid abnormalities of type 2 diabetes mellitus (T2DM) model rats established by high-fat-diet and intraperitoneal injection of STZ, and there exists dose-effect relationship. The completed Youth Found item of the Natural Science Foundation results showed that Wendan decoction could improve the glucolipid metabolic disorder and insulin resistence of the IGT rats which induced by long time high-fat-diet and its mechanism was related to the improvement of liver, skeletal muscle later insulin signaling pathway. This project intends to filter the best compatibility of rhizoma coptidis and Wendan decoction for the treatment of T2DM, make comparisions with Wendan decoction and rhizoma coptidis on their mechanism targets from transcriptomics and proteomics levels, in order to provide experimental basis for the T2DM treatment of Huanglian Wendan decoction and its compatibility, offer research thoughts for the related prescriptions and establish a research platform foundation for traditional Chinese medicine prescription system.

随着超重和肥胖人口的不断攀升,中国已成为世界第一糖尿病大国。课题组前期研究认为国人多坐少动、嗜食肥美的不良生活方式造成“痰湿郁热”病理状态是糖尿病等代谢类疾病的病因病机。《六因条辨》黄连温胆汤是临床常用治疗湿热的方剂,也是课题组有关温胆汤系列研究中的主要方剂,但该方在原书中没有用量的记载,限制了临床应用以及科学研究。本课题在前期研究基础上,在中医理论指导下,完成了温胆汤各组成药物定性定量研究,黄连指纹图谱的建立正在进行中,并复制45%脂肪供能高脂饲料喂养5周,一次性腹腔注射1%STZ30ml/L复制T2DM模型,在此模型基础上,探讨了15g黄连+温胆汤原方、10g黄连+温胆汤原方、5g黄连+温胆汤原方三个不同配伍对该模型的药效学差异,在考虑中医理论、药效、药材成本等多方面因素后,筛选出10g黄连+温胆汤原方作为最佳配伍比例,并就该配伍比例复方与10g黄连进行药效学比较,以期为复方应用及与单味药区别提供实验依据。结果显示,各给药组均可降低模型大鼠空腹血糖、对OGTT各时点血糖异常有改善,而最佳配伍组和黄连单味药还可纠正模型大鼠血脂异常,对模型存在的氧化应激状态具有很好纠正作用,但复方仅表现出优于单味药的趋势,而未见统计学差异。该研究结果将为进一步应用分子生物学及系统生物学技术探讨该方治疗T2DM作用机制及作用靶点奠定基础,为黄连温胆汤及其配伍治疗T2DM提供实验依据,为类方研究提供思路。
