This project takes advantage of the physiological ecology, endocrinology, as well as the molecular biology technology to study how temperature affects serum estradiol, testosterone and gonadosomatic index during the gonad development process, and observe the temperature's effect on gonad development as well as how the female germ cell transfers to the male germ cell in the rice field eel. All of these were based on the temperature's effect on parent breeding, spawning, hatching and fry rearing in the species. And then sample the male and female gonad tissue, transcriptome sequence the extracted RNA, sift the differential genes via expression profile, study the key genes to ensure them regulation effect on sex reversal and clarify the molecular mechanisms in rice field eel. Aiming at the special characteristic of low fecundity and natural sex reversal, the effects of lots of factors, such as nutritional condition, stocking densities and exogenous hormones on controlled artificial multiplying and sex regulation were researched and some achievements were made.According to previous experiments, this project makes use of the temperature, the key factor that affects gonad development and sex differentiation, to explore sex differentiation mechanism, which can clarify the sex differentiation mechanism and provide new method and theory for artificial multiplying technology, then the quantity production can be achieved and the problem of fry's shortage can be solved completely in this species.
