前期研究表明,艾灸可调节UC肠道菌群失衡状态。在此基础上,本研究旨在:采用UC临床研究结合相应动物实验,从体内和体外研究两方面:(1)采用16s RNA基因序列和荧光定量PCR等技术,观察艾灸对UC患者和大鼠肠道菌群多样性、外周和结肠免疫效应因子的含量和表达,以及LPS信号通路上游LPS、LBP及CD14含量、跨膜蛋白TLR4的表达,通路上关键分子,以及下游炎症因子的表达,研究艾灸对UC肠道菌群多样性及LPS信号通路的调节作用,(2)应用蛋白质组学技术,研究艾灸UC血清的主要活性成分,(3)采用细胞培养和激光共聚焦成像等技术,观察艾灸血清单核细胞促炎因子和Th1与Th2型细胞因子释放,以及对LPS介导的炎症因子及炎症信号分子的影响,研究艾灸血清对LPS介导的炎症反应的免疫调节作用,阐释艾灸治疗UC的生物效应机制,完善艾灸治疗UC的临床技术,提高其临床疗效。

Early research has shown that moxibustion adjust intestinal flora imbalance in UC rat. On this basis, the UC clinical research combined with the corresponding experiments on animals, from two aspects in vivo and in vitro, this study aims to: (1) Using 16 s RNA gene sequences and fluorescence quantitative PCR techniques, observation the change of intestinal flora diversity and the content and expression OF peripheral and colon immune effect factors in UC patients and UC rat, and on LPS signal pathway upstream LPS, LBP and CD14 content, the expression of TLR4 transmembrane protein, channel on key molecules, and the expression of inflammatory factors, to explore the effcet of moxibustion on UC intestinal flora diversity and LPS signal pathways, (2) By the application of proteomics technology, study the main active components of moxibustion serum of UC, (3) Using cell culture and laser confocal imaging techniques, observation of proinflammatory factor in mononuclear cells, Th1 and Th2 cytokines released, as well as of the expression of the LPS mediated inflammatory factors and inflammatory signaling molecules in moxibustion serum, to explore the effect of moxibustion serum on the immunoregulatory effects of LPS mediated inflammatory response, explain the mechanism of biological effect of moxibustion and perfect the moxibustion technology in the treatment of UC, improve the clinical curative effect.
