针对每年国内外果品采后损失严重的实际问题,开展果实成熟衰老机制的研究具有重要的理论和实际意义。番茄是研究果实成熟的经典模式植物,课题组前期研究表明一些长链非编码RNA参与番茄果实成熟调控。本项目将在整个番茄基因组水平挖掘重要的番茄果实成熟相关转录因子RIN的靶lncRNA;利用病毒诱导基因沉默技术高通量筛选与果实成熟相关的关键靶lncRNA;通过CRISPR-Cas9等技术敲除靶lncRNA位点,解析关键靶lncRNA在番茄果实成熟中的生物功能;利用双荧光素酶报告基因检测系统探明RIN与关键靶lncRNA启动子的相互作用关系,最终准确而清晰的诠释RIN介导的番茄果实成熟相关lncRNA转录生成调控的分子机制。从新的角度审视和探究番茄果实成熟的调控机理,为果实成熟调控提供新的功能分子元件,为改造番茄果实性状提供新的途径。
英文摘要
Due to serious loss of fruits at home and abroad each year, it has important theoretical and practical significance to launch the research on the mechanism of fruit ripening and senescence. Tomato is a classic model plant for the research in fleshy fruit ripening, our previous study showed that long non-conding RNAs (lncRNAs) are involved in the regulation of tomato fruit ripening. We will scan on target lncRNAs of important tomato fruit ripening-related transcription factor RIN at genome-scale level; carry out high-throughput functional screening of key target lncRNAs using virus induced gene silencing; employ CRISPR-Cas9 technology to knock out the locus of key target lncRNAs; and study the relationship of RIN and promoters of key target lncRNAs with dual-luciferase reporter assay in vivo. Overall, these results will elucidate the transcriptional regulation mechanism of tomato fruit ripening-related lncRNAs by RIN. In short, Our work will try to investigate and explore fruit ripening from another point of view of lncRNA, which will provide new biological resource and new way for regulation on fruit ripening.
