申请人长期工作在心血管外科临床一线,以细胞治疗和心肌保护为主研方向。提出“细胞移植全程预处理”的理念,在“细胞源阶段”根据疾病进程选择细胞去抑制心律失常,在“细胞制备阶段”通过低能激光刺激细胞扩增,在“移植阶段”通过远程缺血或停跳预处理提高移植后细胞存活率,并在临床部分得到转化。在心肌保护领域,提出遗传可能影响先心病外科预后,而多能诱导干细胞是研究合并基因或染色体变异的患儿心肌保护的较好模型。发现影响国人先心病心肌保护效果的基因标记物,并成功构建先心病患儿的诱导多功能干细胞株。迄今在国际性期刊发表第一或通讯作者论著28篇。近5年一作3篇,通讯作者15篇(含共同一作和通讯作者),包括Circ,Eur Heart J等。先后荣获中国青年科技奖、教育部新世纪人才、东方新星奖、中国出生缺陷干预奖以及北京市和教育部科技进步一等奖(排名第二)。本课题致力于先心病细胞治疗和心肌保护研究。

Heart diseases are the main burden of diseases in China. Cell-based therapy had emerged as a promising approach to regenerate the damaged heart. As the chief surgeon of Department of Cardiac Surgery and deputy director of the key lab. for cardiac regenerative medicine, the applicant and his team focused on two key points in cell-based therapy, which including “how to improve the efficacy of cell transplantation” and “how to utilize the induced pluripotent stem cells for myocardial protection for congenital heart disease (CHD)”. The applicant proposed a comprehensive strategy by melding the in vitro and in vivo approaches to enhance the efficacy of cell therapy (J Cell Mol Med 2010), which including cell transplantation during arrested heart ( J Throac Cardiovasc Surg 2008,), laser cell preconditioning (Laser Surg Med 2008), and remote ischemic preconditioning (J Mol Cell Cardiol 2013) . These approaches significantly shorten the time for cell preparation, enhance the cell survival, and thereby improve the efficacy of cell transplantation. In the field of CHD, the applicant proposed that inherited genetic factors also could contribute to the myocardial protection during pediatric cardiac surgery. The applicant identified the genetic prognostic and predictive marker for the Chinese patients undergoing CHD repair(Eur Heart J 2012). Furthermore, He established induced pluripotent stem cells from patients with congenital heart disease and Down syndrome (Stem Cell Res Ther 2012). During the past 5 years, he totally published 18 papers as the leading or corresponding author in the field of heart cell therapy and CHD in international peer-reviewed journals.The current application extents his previous observation and focus on the cardiac regeneration and myocardial protection for CHD.
