胚胎发生启动的分子调控机制是植物生殖生物学的核心问题之一,一直备注学者关注。但由于其研究难度很大,迄今仍是不解之谜。我们的前期工作发现了一个新基因GCD1(gamete cell defective 1),该基因突变后雌配子早期发育正常,惟不能最终成熟。这种没有充分成熟的雌配子可正常受精,但受精产生的合子不能启动胚胎发生。因此推测雌配子成熟时期建立了某种关键的母本机制,这些母本因子对于受精后胚胎发生的启动起着决定性的调控作用。该基因的发现为探讨胚胎发生启动的分子调控机制提供了不可多得的材料和切实的切入点。我们拟以此为基础,通过成熟与不成熟卵细胞转录组的比较,以及GCD1调控途径的分析寻找启动胚胎发生的分子基础,揭示雌配子成熟过程中其功能特化的实质。希望藉此在认识胚胎发生启动的分子机制方面能有所突破。
英文摘要
The molecular mechanism regulating the onset of embryogenesis is one of the core questions in the field of sexual plant reproduction and has been paid great attention for several decades. However, due to technical limitations, it still remains a mystery. In our previous work,a novel gene, gamete cell defective 1(GCD1) was found and confirmed to be responsible for the female gamete final maturation. gcd1 egg cell could be normally fertilized but resulted zygote could not be triggered for embryogenesis. Thus, it offers a unique opportunity to investigate the role of maternal factors established during egg cell maturation in the onset of embryogenesis. We will focus on revealing these necessary maternal factors in this project via comparative transcriptome analysis between mature and immature egg cells and regulatory pathway analysis of GCD1. Hope to enhance our understanding of the molecular mechanism triggering embryogenesis in angiosperm.
