鲤鱼(Cyprinus carpio)是世界上最重要的淡水养殖鱼类之一,是亚欧许多国家的主要养殖品种,在我国的水产养殖中占有极其重要的地位。本研究拟在前期鲤鱼基因组研究工作的基础上,对我国重要鲤鱼种质资源群体开展全基因组尺度的单核苷酸多态性位点遗传变异信息采集,系统掌握这些种质群体的遗传变异信息和种群遗传结构,进而对种质特异性性状开展基因组尺度的遗传基础解析和基因调控机制研究。同时,利用丰富的基因型分析工具,对我国鲤鱼种群开展连锁不平衡分析,构建LD模块,绘制基因组单体型图谱,为下一步开展准确、高效的鲤鱼复杂性状遗传基础解析做好必要的工具储备。

The applicant works on genomics and genetics of major aquaculture species in China, mainly on Cyprinids species which are dominant species in freshwater aquaculture industry. In the past five years, after he joined Chinese Academy of Fishery Sciences in 2009, he worked on the Common Carp Genome Project and developed genome resources and genetic tools of common carp quickly, including the BAC library, BAC-end sequences, BAC-based physical map, high density genetic map, transcriptome resource, and the whole genome sequencing, assembly and annotation of the tetraploidized carp genome. In addition, he also works on the high density 250K SNP array of common carp, and applied it on GWAS studies of common carp. He also worked on genetic and evolutionary study of alkaline stress tolerance mechanisms of Leuciscus walechii, which survives in high alkaline soda lakes. He are also involved in projects that develop genome resources of various aquatic animals with collaborators. In the past five years, he is the principle investigator of multiple national research grants, and is associate editor of " Frontiers in livestock genomics" and editorial board member of PLoS ONE. He has published over 60 peer-reviewed papers inlcuding 42 SCI papers, one book, and one book chapter. Those papers have been cited over 1500 times. He will work on haplotype map constrcution of common carp in China in the project.

本项目利用前期开发和制备的鲤鱼基因组图谱、遗传变异图谱和高通量250K SNP分型芯片,对全球13个代表性鲤鱼品系或地理种群开展高通量SNP基因型分析,采集各种群特异性遗传变异位点信息,进而通过比较基因组技术、全基因组关联分析技术等进行品系特异性性状的遗传基础解析和LD分析,构建这些种质群体的单体型模块,构建和绘制我国鲤鱼基因组单体型图谱;项目还完成了荷包红鲤、黄河鲤、镜鲤全基因组深度测序和参考基因组组装、注释和分析工作,更新和完善了鲤鱼基因组参考序列;利用GWAS和QTL作图等策略,完成鲤鱼生长、性别、鳞被、体型、体色等重要经济性状的遗传基础解析工作;这些研究为在我国主要养殖鲤鱼种群中开展精细的性状解析和基因组辅助育种奠定了数据和工具基础。此外,基于在鲤鱼群体基因组学研究中积累的经验和技术,还先后开展和完成了瓦氏雅罗鱼在极端碳酸盐碱性环境下的基因组适应性进化机制解析,中国花鲈群体遗传学和适应性进化研究等工作。本项目3年来参编专著3部,发表论文17篇,其中SCI论文16篇,一区论文5篇,10分以上论文2篇,申请发明专利4项;项目执行期间,共培养青年骨干2名,博士研究生1名,硕士研究生5名;项目负责人先后获得“国家科技进步二等奖”、“茅以升北京青年科技奖”、“中国水产青年科技奖”、“中国水产科学研究院大渔创新奖”、”中国水产科学研究院科技进步二等奖”等表彰,获得福建省“闽江学者”称号,取得博士生导师资格。
