原生动物纤毛虫形成包囊是一个受基因调控的过程,相应基因的选择性表达是导致细胞形态结构变化的分子机制。本研究在前期工作基础上,拟以冠突伪尾柱虫等易诱导形成包囊的纤毛虫为实验材料,应用RNA-seq技术、RNAi技术、实时荧光定量PCR技术、电镜技术、荧光免疫技术、LC—MS/MS和生物信息学技术,大规模筛选和鉴定纤毛虫胞囊形成相关的基因,从中分析筛选出几个胞囊形成关键基因,多角度研究这些关键基因调控机理。从基因层面探索纤毛虫休眠细胞结构分化及其关键基因参与包囊形成的的分子机理,为揭示真核细胞抵抗逆境的机制、阐明真核细胞与环境相互作用的关系提供重要的基础资料。

Encystment of protoza ciliate is a gene regulation process. The selecting expression of the related genes is molecullar mechanism of cellular morphologic structure changes during ciliate encystment. Based on our previous study, we plan to use ciliates such Pseudourostyla cristata which are easily induced encystment as investigating materials. The genes associating with encystment will be screened and indentified at large scale by using RNA-seq, RNAi, qRT-PCR,electron microscopy, fluorescence immunity, LC—MS/MS and bioinformatics. Among these genes, several key genes associating with encystment will be found. The exact role of these key genes in the encystment will be studied at multi angle. This study explores structure differentiation of ciliate resting cyst and the molecullar mechanism of the key genes participating in encystment at gene level, which provides important basic data for revealling mechanism of eukaryocyte resisting adversity, claryfing interaction relationship between eukaryocyte and environment.
