4-甲基甲卡西酮（4-MMC）， 4-甲基乙卡西酮（4-MEC），亚甲基二氧吡咯戊酮（MDPV）和3,4-亚甲二氧基甲卡西酮（Methylone）是4种近年来流行甚广的合成卡西酮类策划药，对其滥用的鉴定、代谢及机制的研究是当前司法鉴定领域的重要需求和国际热点问题之一。本研究拟利用时下先进的代谢组学和cocktail探针药物法的研究策略，用药物探针技术研究这4种策划药对CYP的抑制酶谱，拟通过GC-MS和LC-HRMS检测药物滥用组和阴性对照组大鼠体液成分的变化，用主成分分析法提取CYP活力改变后的内源性代谢物的特征信息，同时结合病理生理现象、体内分布规律等，解析这些标志物形成的分子机制，阐明这些内源性代谢群组的特征改变与药物滥用的关系及机制，推测合成卡西酮类策划药的特征代谢标志物和体内代谢通路，为司法鉴定实践中合成卡西酮类策划药滥用的鉴定及评价提供新的研究思路和技术储备。

4-Methylmethcathinone(4-MMC), 4-Methylethcathinone(4-MEC), Methylenedioxypyrovalerone(MDPV) and 3,4-Methylenedioxy-N-methylcathinone(Methylone) are four kinds of synthetic cathinones which are very popular in recent years. Identification of these abused drugs and their metabolic and mechanism research is one of the most important international issues in current judicial appraisal field. This study intends to use the advanced metabolomics and cocktail probe drugs research strategy to study the influence of 4-MMC, 4-MEC, MDPV and Methylone on CYP enzyme inhibition spectrum. Based on GC-MS and LC-HRMS, we want to detect the metabolic changes after abuse of 4-MMC, 4-MEC, MDPV and Methylone. The feature endogenous metabolites information will be extracted using principal component analysis. Combined with pathophysiological phenomenon and vivo distribution, etc., we will study the molecular mechanisms of these metabolic markers and find the relationship between 4-MMC, 4-MEC, MDPV and Methylone abuse and endogenous metabolites changes. Besides, vivo metabolic pathways of 4-MMC, 4-MEC, MDPV and Methylone will be find. The initial result will be used to provide reference and technical support to the identification of the abuse of synthetic cathinones.
