针刺手法的补泻效应已经得到临床和实验验证,然而效应机制并不明晰。结缔组织成纤维细胞是将机械信号转化为生物学信号的关键环节。在课题组以往研究基础上,本研究采用正交试验设计和析因分析方法,在曲池穴选取不同频率、用力方向和刺激时间的3因素3水平治疗条件组合的9种针刺手法方案治疗内毒素致热家兔,观测穴区成纤维细胞形态,成纤维细胞合成和分泌嘌呤受体信号分子ATP、机械信号转导因子PGE2、NO和细胞外基质影响因子TGF-β1、MMP-1的功能,以及家兔肛温、内源性致热因子IL-1β、IL-6、TGF-β和下丘脑中枢性发热介质PGE2、cAMP、AVP、α-MSN的含量。旨在探讨影响提插手法补泻效应的主次因素及各个因素之间的交互作用,分析穴区成纤维细胞形态功能改变与靶器官补泻效应的相关性,从细胞生物力学角度揭示成纤维细胞在针刺手法力学信号转换中的重要作用,为临床针刺补泻手法的应用和传承提供实验依据。

Reinforcing and reducing effect of acupuncture manipulations has been verified by clinical and experimental researches, while its mechanism is not clear. Fibroblast is a key link in the transforming process from mechanical signal into biological signal. Based on previous studies, this study adopts orthogonal design and factorial analysis, combining 3 factors(frequency, direction and stimulation duration) and 3 levels into 9 kinds of acupuncture manipulations which are applied in the point LI11 to treat fever rabbits. Fibroblast shape in acupoint area, Purine receptor signaling molecule ATP, mechanical signal transduction factors PGE2 and NO and extracellular matrix factor TGF-β1 and MMP-1 which are produced and synthesized by fibroblast, anal temperature, endogenous pyrogen IL-1β,IL-6,TGF-β in serum and central heating medium PGE2,cAMP,AVP and α-MSN in hypothalamus are observed. The purpose of this project is to find out primary and secondary factors which influence reinforcing and reducing effect and the interaction between various factors, analyze relationship between fibroblasts in acupoint area and reinforcing and reducing effect on target organs, reveal the important role of fibroblasts in mechanical signal transformation process of acupuncture manipulation, and provide experimental basis for clinical application and inheritance of acupuncture manipulations.
