大气细颗粒物(PM2.5)污染已成为危害人民健康的重要因素,是急需解决的国家重大需求。项目组前期采用鼻腔滴注PM2.5悬液的方法建立了PM2.5致小鼠肺损伤模型,发现白及提取物对PM2.5引起的小鼠肺损伤具有保护作用,但具体作用机制未知。PM2.5对机体最典型的损伤机制是发生于呼吸道的炎症反应,IL-1β活化分泌对PM2.5损伤肺的发生发展具有重要作用。我们在明确白及活性成分的基础上,进一步明确筛选出的白及抑制IL1-β分泌药效成分对PM2.5引起的小鼠肺损伤是否具有保护和预保护作用,重点从TLR4/NF-κB通路、ROS 及炎性小体等关键靶点阐明白及药效成分抑制IL1-β分泌的分子机理。本项目在中医“治未病”、“既病防变”思想指导下,开展抗PM2.5中药药效成分及作用机理研究,对保障人民身体健康、提升人民生活质量、缓解社会矛盾等均有重要意义。

Atmospheric fine particulate matter (PM2.5) pollution has endangered people's health,which has become an urgent need to address the major needs of the country. In our earlier study, PM2.5 induced lung injury mouse model was established by intranasal instillation of PM2.5 in mice. We found that Bletilla striata (Thunb.) Reiehb.f extract had protective effects on lung injury induced by PM2.5 in mice. We will further investigate the mechanism. the most typical damage mechanism of PM2.5 on the body is the inflammation of the respiratory tract and the activation secretion of IL-1β played an important role in the occurrence and development of PM2.5 injured lung. On the basis of the active components of Bletilla striata (Thunb.) Reiehb.f, we will specifically screen out the active ingredients which inhibit the secretion of IL-1β to see whether they have the function of protection and pre-protection on lung injury induced by PM2.5 in mice. we will focus on clarifying the molecular mechanism of effective constituents of Bletilla striata (Thunb.) Reiehb.f which inhibit the secretion of IL-1β from the key target such as TLR4/NF-κB pathway, ROS and inflammatory corpuscle. This study is under the guidance of TCM such as preventive treatment of disease and prevention to carry out the research on the composition and action mechanism of Chinese medicine anti PM2.5, which is of great significance in improving health and quality of life and at the same time alleviating social contradictions.
