蛴螬是一种重要的地下害虫,在我国为害的蛴螬有100多种,作物地下部分损失的86℅是由蛴螬造成的,并且其为害程度有日益严重的趋势,目前尚缺乏有效防治措施。理解“植物-害虫-害虫病原” 间相互关系是探索害虫可持续控制途径的重要基础,在基因水平研究三者间相互作用关系可以提供害虫防治的新思路。本研究利用实验室之前获得的蛴螬取食主要寄主花生及其重要病原菌苏云金芽胞杆菌前后,Bt、蛴螬、花生三者的转录组数据,在基因水平相对三者相互应答情况进行深入分析;结合目前其他可用的数据库资源(例如,KEGG),在基因水平对三者相互作用过程涉及的信号途径网络进行挖掘分析;进一步建立界面友好的数据库,为进一步理清“花生-蛴螬-Bt”相互作用的研究建立生物信息支持平台,也为进一步探索通过基因干预对蛴螬进行防控方法研究提供依据。

White grubs is an important underground pests, in China, there are more than 100 damaging grub species and causes 86 percentages loss of crops underground parts; its harm degree has a growing trend and there still lack of effective prevention and control measure now. Understanding the relationship between "Host plants - Insects - Pathogens" is an important foundation to explore the sustainable pest control measure while investigating the relationship between "Host plants - Insects - Pathogens" in genetic level may provide new ideas for pest control. In this project, we use the Holotrichia parallela, Arachis hypogaea and Bt’s transcriptome data that obtained from the samples collected before or after H. parallela ingest the A. hypogaea or Bt, to analysis the relationship between "Bt - White grub - Peanuts" at the genetic level; Combining with other available database resources (for example, KEGG), this research will mining and analysis the signaling pathways involved in the process of interaction between "Bt - White grub - Peanuts"; furthermore, this work will establish database with friendly interface and provide the biological information supporting platform for the further understanding of interaction between "Peanuts - Grubs - Bt", and it also will provide the basis for the development of prevention and control methods to control grubs using gene interference measure.

蛴螬是一种重要的地下害虫,在我国为害的蛴螬有100多种,作物地下部分损失的86℅是由蛴螬造成的,并且其为害程度有日益严重的趋势,目前尚缺乏有效防治措施。理解“植物-害虫-害虫病原” 间相互关系是探索害虫可持续控制途径的重要基础,在基因水平研究三者间相互作用关系可以提供害虫防治的新思路。本研究测定了花生基因组数据并汇集成PeanutDB花生数据库以及关于陆生植物与绿藻类植物包括花生等植物及基因组的数据库PlantOrDB。测定了多个Bt菌株的基因组并对14株已有完成图数据的Bt菌株进行泛基因组分析,建立了关于Bt泛基因组的数据库BTBIDB。测定并组装了暗黑鳃金龟、大黑鳃金龟和白星花金龟中肠组织的转录组并对预测蛋白进行注释,鉴定 Bt蛋白与蛴螬中肠的相互作用。进一步评估了微生物群落在“花生-蛴螬-Bt”这三者相互作用网络中的作用,为进一步探索通过基因干预对蛴螬进行防控方法研究提供依据。
