申请人长期从事病毒和生物大分子的冷冻电子显微学研究,主要成果有:1)获得了30nm染色质纤维(Science 2014)及染色质重塑复合物Tip49a/b的高分辨率结构,在染色质高级结构及调控这一重大科学疑难问题中取得了突破性进展;2)解析了目前唯一一个病毒转录前后状态的全原子分辨率结构模型(PNAS 2011, PNAS 2012),揭示了双链RNA病毒的转录机制以及病毒新生RNA的加帽和流出通道;3)在艾滋病毒、手足口病病毒等重要致病病毒及疫苗的结构学评价上取得了重要进展。近年来,申请人在包括Nature, Science, PNAS, PLoS Pathog., J. Virol.等知名期刊上发表论文近30篇;近五年内共发表学术论文14篇,其中通讯或共同通讯作者7篇(含Science一篇,PNAS两篇);所有论文在最近5年中的SCI总引906次,他引823次,单篇最高他引215次。

The applicant has been working on the structural studies of virus and biomacromolecules using cryo-electron microscopy (cryo-EM) analysis for a long period of time,and made noticeable achievements: 1) Acquired the high resolution structures of 30nm chromatin fiber (Science 2014) and chromatin remodeling complex Tip49a/b, which is a breakthrough in the research field of chromatin structure and epigenetic regulation; 2) Built the first, currently the only, atomic model of a cypovirus in both the native state (PNAS 2011) and the transcribing state (PNAS 2012), which provides critical insights into the mechanism of transcription of dsRNA virus and the capping and transportation channel of the nascent mRNA during the transcription process; 3) Made significant progress on the structure analysis and evalution of the virus like particle (VLP) vaccines, as well as the corresponding pathogenic viruses, against AIDS and Hand, Foot, Mouth Disease (HFMD). Near 30 papers have been published by the applicant in pretigeous journals, including Nature, Science and PNAS, J Virol., etc. In the past 5 years, the applicant has published 14 papers, among of which 7 as the correspondence or co-correspondece (including Science 2014, PNAS 2011, PNAS 2012). According to the scientific citation index reports, publications by the applicant have been cited 906 times in total during the past five years. The highest non-self citation number for one single publication is 215.
