药对配伍研究是复方配伍规律研究的基础和重要的切入点之一,但目前药对配伍规律的研究尚缺乏点面结合较系统的方法学,阻碍了中药配伍研究取得突破性进展。“组合微透析技术”是采用微透析技术进行多位点同步取样,并将探针与多维液相色谱仪、多级质谱、生物信号采集处理系统等进行在线连接,建立微透析技术与色谱、质谱、核磁共振联用的复合技术,微透析技术与药代动力学、中药药理学等多学科结合的方法,对药对或复方来源的“吸收组分群”及内源性物质的动态变化进行研究。有效实现了活体、在线、药动-药效-药分的同步结合研究,与中医药理论整体观、动态观相吻合。故本课题以附子-大黄药对为研究载体,基于“药性-药对-证型”的研究思路,利用符合中医证候的模型大鼠,采用与中医药理论整体观、动态观相吻合的组合微透析技术进行药对配伍规律的研究,以揭示中药药对配伍规律的现代科学本质,并为现代方剂配伍提供新的思路和方法

Drug pair is the basis of the study of compound collocation, but nowadays the research on collocation rules is still lack of integrate system methodology,which has hindered the breakthrough of traditional Chinese medicine research.The project is based on "YAOXING-YAODUI-ZHENGXING" axis, with a representative of the "aconiti-rhubarb" drugs as the research object, using proper syndromes model and physiological rat to living, online, medicine efficacy and traditional Chinese medicine theory and the overall concept, dynamic view coincide with the combination of microdialysis technology as the core, with "material foundation and mechanism" for the study of the carrier, the integrated use of interdisciplinary, namely in with "absorb" as the breakthrough point, to grasp the medicine to "absorb the group -dividing" information, based on study of TCM syndrome type, implementing the drug analysis-pharmacokinetics- pharmacodynamics (PA-PK-PD) 3d synchronous experimental research, blood inside body, at the same time the liver, the kidney and so on many sites myocardial, components (group) and endogenous substances and pharmacodynamics the dynamic change of the correlation between changes in the study. Integrated absorption, PA-PK-PD, clarified systematically "aconiti-rhubarb" medicine material base for effect and action mechanism, revealed "aconiti-rhubarb" medicine in building a set of rules for scientific evaluation system of medicine lays the foundation, to provide a theoretical basis for clinical formula.

本项目综合运用药代动力学、组织药理学及中医基础理论等多学科交叉,结合“组合微透析技术”,研究阐明“附子-大黄”药对配伍的现代内涵,探讨中药药对配伍规律,以期为临床药对及复方配伍提供实验依据。采用二次评价方法(生理生化指标、以方测证)建立阳虚便秘证候模型;采用血液微透析技术进行药对主要组分的药动学研究,观察组分在模型大鼠体内的吸收代谢过程;采用免疫组化、RT-PCR等技术检测相关药效学指标,综合分析“附子-大黄”药对的物质基础和配伍机制。
