本研究计划通过收集、处理和保存去世后捐献的人脑组织样本,建立规范化的中国人脑组织资源库,参照国际通用标准对脑组织样本进行神经病理学检查和分类,构建包括临床和病理学资料在内的数据库,并建立规范的样本分享与合作研究机制。在中国人脑组织库的基础上,开展对脑衰老和阿尔兹海默病发病机制的研究。通过对多年龄组的人脑组织样品的检测和分析,利用形态学、分子生物学和高通量组学分析等技术手段,揭示人脑组织在衰老和阿尔兹海默病发展过程中的生理和病理学变化,为未来的诊疗提供实验依据。通过鉴定在以上过程中主要发生变化的蛋白分子,并采用生物信息学分析和分子生物学等手段研究其生物学功能,以揭示其在阿尔兹海默病发病机制中扮演的角色,为寻找新的诊断治疗靶点及药物提供思路和理论基础。
英文摘要
We propose to construct a standardized human brain bank by collecting, processing and storing postmortem human brain tissue via a willed donation program in China. Neuropathological analysis and classifications for human brain tissue will be performed according to internationally recognized standards. A database including clinical and neuropathological data will be built, with proper tissue staring and collaborative research mechanisms. Based on the human brain bank, we will study the mechanisms of human brain aging and Alzheimer’s disease on brain tissue from different age groups using a combination of morphological, molecular biological and high-throughput proteomic techniques. The functions of key protein molecules discovered during the above process will be verified and further studied using bioinformatics analysis and molecular biological methods to reveal their roles in Alzheimer’s disease. This project may provide novel strategies and theoretic basis for the diagnosis and treatment of Alzheimer’s disease.
