In order to analysis Pseudopeziza medicaginis(Lib.)Sacc infecting alfalfa this cytological event from protein level, based on the fully understanding of the biological characteristics and infection process of P. medicaginis, physiological metabolism, pathogenic differential proteomics, gene cloning and expression of related proteins genes during P. medicaginis the growth and its infection of alfalfa leaves will be systematically studied by the comprehensive use of two-dimensional electrophoresis (2-DE), mass spectrometry (MALDI-TOF-TOF/MS) and physiological method. Differential protein expression changes will be analyzed, the key differential expression proteins related to pathogenic process will be identified and their expression pattern will be revealed; main physiological changes of alfalfa responses to P. medicaginis infection will be analyzed, combined with the physiological index, physiological functions of key proteins involved in host tissue pathological changes will also be analyzed; function modules or biological functions of the key differential proteins will be identified by bioinformatics analysis; part of differential protein genes related to the growth and pathogenicity will be cloned, bioinformatics analysis will be studied. These results will lay the foundations for revealing the molecular mechanism of P. medicaginis pathogenicity and for investigating the unique infection strategy of P. medicaginis successfully infects the host.
