本项目以解决影响肝炎和肝纤维化疾病进程免疫调控机制中的重大科学问题为导向,利用我实验室已建立的技术平台和免疫细胞新亚群研究的技术体系结合双光子显微镜活体成像可视化技术平台,在构建ConA诱导的小鼠肝炎模型和免疫性肝炎肝纤维化疾病模型的基础上研究和分析肝脏内巨噬细胞的表型和功能变化,发现并鉴定出与肝炎和肝纤维化进程密切相关的新细胞亚群,并深入研究其在肝脏炎性损伤、肝纤维化和去纤维化中的作用,分析该细胞亚群与肝炎和肝纤维化病程进展的关系,以及该细胞亚群具体的作用机制和分子调控网络,并从中鉴定出影响肝脏巨噬细胞表型和功能转变的关键分子和相应的药物作用靶点。
英文摘要
The project aims to solve common frontier scientific issues for the immune regulatory mechanisms in hepatitis and liver fibrosis disease, particularly, the analysis of cellular and molecular immune regulatory network under physiological or pathological conditions in liver. We have been established technology platforms to find and identify the new immune cell subsets, together with imaging visualization technology in vivo through two-photon microscopy. To discover and identify the new cell subsets and their functions in hepatitis and liver fibrosis progression, we will explore the phenotypic and functional changes in liver macrophages in the mouse disease models. Of important, we devote ourselves to find the new cellular mechanism underlying the hepatitis and liver fibrosis and the new key molecules as potential drug targets.
