传统中医理论认为肝肾不足、湿浊瘀血阻络是糖尿病视网膜病变(DR)的基本病机，临证治以补益肝肾、祛浊化瘀、活血通络。以法组方通络驻景丸用于治疗DR，颇具中医特色与优势且疗效确切，但其作用机制不清，生物学基础不明。组蛋白甲基化在代谢记忆形成过程中发挥了关键作用，前期研究发现通络驻景丸治疗DR的作用机制与代谢记忆密切相关，因此提出假说认为通络驻景丸可通过干预组蛋白甲基化，从而阻断代谢记忆过程，发挥治疗DR的作用。本项目通过构建体内外实验模型，采用CHIP, qRT-PCR, Western blot,流式细胞等技术，检测SET7, LSD1, SUV420H2, H3K4, H3K9, H4K20, sod2, IL-6等蛋白和mRNA表达及生物学效应指标，从表观遗传学角度探讨通络驻景丸干预代谢记忆的作用机制，对揭示其作用靶点和作用规律具有重要意义，同时为中医药防治DR的机制研究提供新的思路。

Traditional Chinese medical theory holds that deficiency of liver and kidney, ecchymosis, sputum is the main pathogenesis of diabetic retinopathy (DR). Based on this, treating DR by the theory of tonifying the liver and kidney, eliminating dampness and removing blood stasis and promoting blood circulation to remove meridian obstruction, Tongluo Zhujing pills have obvious advantages and characteristics. However, the research on its mechanism and biological base does not go further enough. Histone methylation plays a significant role in the formation of metabolic memory which is closely related to the mechanism that Tongluo Zhujing pills treat DR in preliminary study. Accordingly, we put forward the hypothesis that Tongluo Zhujing pills can block metabolic memory and treat DR through effecting the histone methylation. Building models in vivo and in vitro, we test the protein and mRNA expression of SET7, LSD1, SUV420H2, H3K4, H3K9, H4K20, sod2, IL-6, et al and biological effect index by siRNA, CHIP, qRT-PCR, western blot, flow cytometry and other technology. Our research explores the mechanism that Tongluo Zhujing pills intervene metabolic memory from the perspective of epigenetics. It is of great importance in the therapeutic targets and laws, providing new ideas for the research on mechanism of DR prevention and treatment using TCM.
