未绝经女性较同龄男性冠心病发病率低,且不易出现心肌缺血再灌注损伤(MIRI),绝经后则出现相反结果,因缺乏雌激素所致。雌激素替代疗法对绝经女性冠心病及MIRI具有治疗作用,但久用雌激素又增加罹患妇科肿瘤的危险,而植物雌激素无此副作用,是一类有前途的雌激素替代物。二仙(仙茅、仙灵脾)含有较多植物雌激素,其补肾助阳之功效恰能纠正绝经后冠心病“病位在心,但本源为肾阳气虚衰”之病机,而且本课题前期研究发现二仙能降低MIRI大鼠模型的心梗面积。故本实验利用HPLC和GC/MS/DS系统测定二仙内所含的植物雌激素,利用老年雌性大鼠MIRI模型和心肌细胞缺氧复氧模型,在体离体相结合,探讨二仙及其配伍预处理的抗凋亡、抗炎、抗氧化的心脏保护作用及与GPER受体信号通路的关系,同时研究其有效成分与药效学关系。预期可为开发出新的植物源性雌激素替代品提供实验依据,又可为从肾论治绝经后冠心病中医理论提供实验基础。

Many epidemiological studies and experimental observation had demonstrated that premenopausal females have a lower incidence of cardiovascular disease compared with their male counterparts,and premenopausal females were not prone to ischemia reperfusion injury than postmenopausal. The phenomenon was caused by estrogen. Estrogen replacement therapy has better curative effect on preventing myocardial ischemia reperfusion injury, but Chronic exposure to estrogen eventually will lead to the risk of gynecologic malignant tumors,but phytoestrogen without the effects. Rhizoma curculiginis and Herba Epimedii contains a lot of phytoestrogen and have estrogenic effects.According to the viewpoint of Chinese Medicine,coronary disease of postmenopausal women was a heart disease, but the origin was ShenXu.Tonifying Shen Yang of Er-Xian coincided with the pathogenesis, so Er-Xian might be as an alternative to estrogen. Furthermore, the previous study found that Er-Xian can reduce myocardial infarction area in the MIRI model. Based on this,the protective effects of Er-Xian the on MIRI and relationship with G protein coupled estrogen receptor (GPER) signaling pathway were explored in the study.Firstly,the goal of the present study is exploring material basis of Curculigo orchioides and Herba Epimedii and their pharmacodynamics by HPLC (High-Performance Liquid Chromatography) and GC/MS/DS analysis.Secondly, the prevention effects of Er-Xian on myocardium ischemia-reperfusion injury of aged female rats through in vivo and in vitro will be explored in the study.At the same time,the relationship the mechanism of Er-Xian protective effects with G-protein coupled estrogen receptor(GPER) and its related signal pathway will be explored too. The expected results of the study not only for finding drugs of plant origin estrogen replacement lay the initial foundation,but also for treatment from the Shen of postmenopausal coronary heart disease provide experimental basis.
