已有的工作证实 “健脾益肾,活血通络”为原则创立的 “芪归益肾方”具有保护肾小球滤过膜和减轻肾小管上皮细胞转分化的作用。随着研究的深入,课题组基于前期工作基础提出应用改良的中药方剂调控功能蛋白表达,干预相关细胞信号通路可以有效减轻肾脏纤维化从而治疗慢性肾脏病。 本研究拟通过“调控功能蛋白SIRT1表达,影响NF-KB相关信号通路,保护肾功能”的视角,借助具有自主知识产权的 “芪归益肾方”探讨改良中药方剂治疗慢性肾脏病的核心机制。研究借助典型的动物和细胞模型,探讨改良的中药方剂通过调控SIRT1蛋白表达进而影响基因调控、细胞外基质代谢和免疫相关的炎症反应等作用阐明中医药在慢性肾脏病领域的应用价值。研究采用蛋白组学、免疫沉淀、蛋白免疫印记、液质联用和基因芯片等技术手段阐明中药方剂延缓肾脏纤维化的物质基础和作用机制。

Our work had confirmed that the prescription of ‘qi gui yi shen decotion(QGYS)’ could protect the glomerular filtration membrane and decrease renal tubular epithelial cell transdifferentiation. QGYS base on the principle of " Strengthening spleen and nourishing kidney, promoting blood circulation and removing obstruction in channels " . With the deepening of the research, we speculate these phenomenon relate to the level of sirt1. We want to find the mechanism among the level of sirt1, the signaling pathway of NF-KB and renal function. With the advanced technology of pharmacology and pharmaceutical engineering, we would do a series of experiments in vivo and in vitro to observe the change of sirtuins and the reactions such as the metabolism of extracellular matrix, immune inflammatory reaction and the role of the cell cycle genes, etc. We would do more experiments to confirm the innate relation of these phenomenon. We will use some of the molecular biological methods including proteomics, Immunoprecipitation , western blotting, high-performance liquid chromatography-mass spectrometry and gene chip and so on, to clarify the mechanisms of delaying renal fibrosis and the material basis of Chinese herbs.
