青光眼是全球首位不可逆性致盲眼病,滤过性手术(GFS)是目前治疗青光眼的主要手段。但GFS术后滤过道瘢痕化导致术后高失败率,一直是青光眼研究领域未能攻克的重要难题。中药复方青光安颗粒在已有的研究中显示,具有减轻GFS术后炎症反应,减少结膜下瘢痕面积和成纤维细胞增殖,从而预防术后眼压回升的作用。它抗瘢痕作用的具体机制和实现途径是值得研究的课题。自噬已经被证实在维持细胞稳定及适应应激方面起重要的作用,但是过度的自噬却会对细胞死亡,特别是凋亡有促进作用。中医药诱导自噬的研究在近几年已屡见报道。本项目通过培养人Tenon’s 囊成纤维细胞(HTFs)进行体外试验和构建动物模型进行在体验证,探讨青光安颗粒剂是否诱导了抗青光眼术后HTFs的自噬性死亡。本课题是对项目组已有研究结果的有力补充,能为青光安作用机制的进一步研究开拓新的领域,并提供实验依据和理论依据,进而有力地推动新型抗滤过道瘢痕药物的开发。

Glaucoma is the first irreversible disease causing blind in the global. Filtration surgery(GFS) currently is the main means of curing glaucoma, but bleb scar formation after GFS enabling failure rate to reach up to 15% after one year of surgery, while failure rate after five year of surgery is up to 30%. Effective and safe anti-scarring drugs have not been discovered until now. Chinese herbal compound QingGuangAn granula has already been displayed in previous studies and is equipped with functions of alleviating GFS postoperative inflammatory response, reducing scarring area under conjunctiva and proliferation of fibroblast, so as to prevent from postoperative intraocular pressure recovery. The specific mechanism and realization approach of anti-scarring functions are deserved to study. It is proved that autophagy plays an important role on cell stability and adapting to stress, but excessive autophagy will accelerate cell death, especially for apoptosis. The study on TCM inducing autophagy has already been reported repeatedly in recent years. By cultivating human Tenon’s fibroblast (HTFs), the project conducts extracellular experiments and constructs animal models to verify, discusses whether QingGuangAn granula induces autophagy death of HTFs after glaucoma surgery. The topic is a powerful supplement to known research results of the project team, expands a new field for further studying QingGuangAn mechanism, provides experimental basis and theoretical basis, and then powerfully promotes development of new-type anti-scarring drugs.
