女性围绝经期卵巢功能衰退,雌激素分泌减少所致的血管收缩舒张功能失调是导致女性围绝经期高血压的重要病理基础。鉴于应用雌激素的副作用,从中药中开发安全有效的植物雌激素极具应用价值。本申报项目在既往杜仲雌激素样活性成分围绝经期综合征心血管保护作用系列研究基础之上,基于雌激素对血管收缩舒张功能调控的基因组效应和非基因组效应,首先应用大鼠离体胸主动脉环实验,原代大鼠心脏微血管内皮细胞和胸主动脉平滑肌细胞研究杜仲雌激素样活性成分调控血管收缩舒张功能的分子机制;而后采用整体动物实验,研究杜仲雌激素样活性成分预防去势雌性SD大鼠血压升高的作用。为女性围绝经期杜仲及其提取物的合理应用提供科学依据,促进中医药理论的继承、发展及创新。

Vasomotor dysfunction caused by estrogen secretion decline is an important pathologic basis of hypertension in perimenopausal women. In view of the side effects of the application of estrogen, more safe and effective phytoestrogens development from the traditional Chinese medicine has a good application value. The application is based on the research of the cardiovascular protection effects of estrogen-like activity compounds from Eucommia ulmoides Oliv. in perimenopausal women and the genome effect and non genome effect of estrogen on the regulation of vasomotor function. Firstly, we use thoracic aortic rings, cardiac microvascular endothelial cells and thoracic aortic smooth muscle cells from rats to study the molecular mechanism about the regulation effects of the estrogen-like activity compounds on vasomotor function. Secondly, we use ovariectomized female rats to study the anti-hypertension effect of the estrogen-like activity compounds. The experimental results will be a scientific basis for the reasonable application of Eucommia ulmoides Oliv. and its extract, and promote the inheritance, development and innovation of TCM theory.
