杨树转录因子ERF76基因属于ERF转录因子基因家族,是一个受胁迫诱导表达的基因。为探明转录因子ERF76基因在杨树生长发育和抗旱耐盐胁迫过程中的生物功能,本研究拟进行以下四个方面的研究:首先,培育转录因子ERF76基因过量表达和抑制表达的杨树转基因株系,分析其形态特征、生理特征和抗旱耐盐能力的变化;其次,利用Chip-Seq分析与杨树转录因子ERF76结合的DNA序列特征,结合转录组测序结果鉴定杨树转录因子ERF76调控的下游靶基因;第三,研究杨树转录因子ERF76与其下游靶基因顺式作用元件的结合能力及与其他蛋白的相互作用,明确其转录激活能力和转录调控功能;第四,利用iTRAQ方法定量分析转基因株系的差异表达蛋白质,与转录组测序确定的下游靶基因互相印证。通过以上不同层次的研究,阐明杨树转录因子ERF76在杨树生长发育和抗逆胁迫过程中的作用机制及分子生物学功能。

Transcription factor ERF76 gene from poplar belongs to ERF transcription factor gene family, and its expression is induced by stress. For the understanding of the molecular biological of transcription factor ERF76 gene function in poplar, the studies will be performed as following. First, to cultivate the transgenic poplar of overexpression and inhibit expression of transcription factor ERF76 gene, and analyze the changes of the morphological trait, physiological characteristics, and the resistant ability against drought and salt; Second, to analyze the DNA sequences bound by transcription factors ERF76 using Chip-Seq method, to identify the downstream target gene regulated by transcriptional factor ERF76 with poplar transcriptome sequencing, to analyze the metabolic processes involved in or regulated by these gene products; Third, to detect the binding capacity of transcription factor ERF76 protein with the GCC-box, DRE and CRT and other cis acting elements, and the interaction between the transcription factor ERF76 and other proteins, to clear its transcriptional regulation functions and the effects of different protein on transcriptional activation ability; Fourth, to detect the differentially expressed proteins between transgenic lines and non transgenic lines using iTRAQ method, to confirm each other with transcriptome sequencing results. Through the different levels of the study, to clarify the mechanism of transcription factors ERF76 in the process of growth, development and stress resistance in poplar, to clarify the molecular biology of the transcription factor ERF76 function.
